Correlation Between SCD40L,NF-?B,PTX3 And Cerebral Infarction,CD40 Gene Rs1535045 Single Nucleotide Polymorphism

Objective:1.To investigate the relationship between the expression levels of soluble CD40 ligand(s CD40L),nuclear factor-?B(NF-?B),human pentagoxin 3(PTX3)in human peripheral blood and cerebral infarction.And to explore the correlation of s CD40 L,NF-?B,PTX3 in human peripheral blood.2.To observe the association between CD40 gene rs1535045 single nucleotide polymorphisms(SNP)and s CD40 L,NF-?B and PTX3 and to explore the possible inflammatory signaling pathway of rs1535045 SNP in arteriosclerosis.Methods:90 patients with acute cerebral infarction were selected as the case group,and 85 healthy persons were selected as the control group.The rs1535045 SNP genotypes were determined by Taq Man probe technique.The plasma levels of s CD40 L and PTX3 were detected by enzyme-linked immunosorbent assay(ELISA).The positive rate of NF-?B in human peripheral blood mononuclear cells(PBMCs)was detected by flow cytometry(FCM).SPSS 20.0 software was used to analysis the data of expression of s CD40 L,PTX3 and NF-?Bp65 in the two groups,and the correlation belong the three inflammatory factors and the relationship between rs1535045 SNP.Results:1.Case group:the plasma concentration of s CD40 L and PTX3 were(64.952±19.474)pg / m L and(1.957±0.599)ng / m L,respectively;the positive rate of NF-?B in PBMCs was(30.883±12.748)%.Control group:the plasma concentration of s CD40 L and PTX3were(59.176 ±16.698)pg / m L and(1.737±0.632)ng / m L,respectively;and the positive rate of NF-?B in PBMCs was(18.082±8.674)%.The plasma concentration of s CD40L?PTX3 and NF-?B positive rate in case group were significantly higher than those in control group(P <0.05).2.In all samples: there was a positive correlation between s CD40 L plasma level and NF-?B positive rate(r = 0.485,P <0.001);there was a positive correlation between s CD40 L and PTX3 plasma levels(r = 0.251,P = 0.001);and there was a positive correlation between PTX3 and NF-?B expression level(r = 0.301,P <0.001).In the case group,there was a positive correlation between s CD40 L plasma level and NF-?B positive rate(r =0.764,P <0.001);there was a positive correlation between s CD40 L and PTX3 plasma level(r = 0.445,P <0.05);and there was a positive correlation between NF-?B and PTX3expression(r = 0.384,P <0.05).In the control group,there was no correlation between s CD40 L plasma level and NF-?B positive rate,P = 0.716.There was no correlation between s CD40 L and PTX3 plasma level,p = 0.867;NF-?B positive There was a correlation between the expression rate and PTX3 plasma level,P = 0.558.3.The the plasma concentration of s CD40 L in CD40 gene rs1535045 CC genotype,CT genotype and TT genotype were(59.555±18.070)pg / ml,(63.453±17.193)pg / ml,(65.278±23.545)pg / ml,respectively;NF-?B p65 was(22.671±11.183)%,(25.365±12.287)%,(28.400±17.757)%,respectively;plasma PTX3 concentration was(1.810±0.610)ng / ml,(1.863±0.609)ng / ml,(1.927±0.745)ng / ml,respectively.The differences between these three genotypes were not statistically significant,but there were concentration gradients.There was no significant difference in plasma s CD40 L concentration,PTX3 concentration and NF-?B p65 positive rate between the three genotypes whether in the cerebral infarction group and the control group(P> 0.05).There was no significant difference in the distribution of plasma s CD40 L concentration,PTX3 concentration and NF-?B p65 positive rate in the LAA group(P> 0.05),but there were concentration gradients aslo.Conclusion:1.The levels of s CD40 L,PTX3 and NF-?B in patients with acute cerebral infarction were significantly increased,and there were positive correlations between two inflammatory factors respectively.2.We don't think there is a relationship between rs1535045 SNP and s CD40 L,NF-?B and PTX3 expression levels.
