Aims:To research the change of serum miR-126 and MMP-13 expression of patients with atherosclerosis cerebral infarction.To explore the potential role of miR-126 and MMP-13 as a novel biomarker to predict and early diagnose atherosclerosis cerebral infarction.Method: Collecting serum specimens of patients with atherosclerosis cerebral infarction(n=40),Non atherosclerosis cerebral infarction(n=30),Asymptomatic cerebral atherosderosis(n=32)and gender,age matched control individuals(n=35).the expression of miR-126 and MMP-13 in serum were respectively measured by real-time quantitative PCR(RT-qPCR)and enzyme linked immunosorbent assay(Elisa).Analyzing the difference of the two indexs between each groups.Exploring the relationship between miR-126,MMP-13 and NIHSS,ADL of patients with atherosclerosis cerebral infarction.Result:1.We found that 3'UTR of gene that code MMP-13 have miR-126 binding site.2.The serum miR-126 level of ACI group and ACA group were lower than control individuals and NACI group(P<0.01),The serum miR-126 level of ACI group was lower than ACA group(P<0.01),There was no significant difference on serum miR-126 level between NACI group and control group(P>0.05).3.The serum MMP-13 level of ACI group,NACI group,ACA group were significant higher than nomal group(P<0.01),The levels of miR-126 of ACI group was significantly higher than NACI group and ACA group(P<0.01).4.The ACI group was divided into serious condition group(NIHSS>4)and milder condition group(NIHSS?4).The serum miR-126 level of serious condition group was significantly lower than milder condition group(P<0.01).Pearson's correlation analysis showed that the serum miR-126 level of ACI group was negatively correlated with NIHSS(r=-0.691,P<0.01).The serum miR-126 level of ACI group was positively correlated with ADL(r=0.923,P<0.01).5.The serum MMP-13 level of serious condition group was significantly higher than milder condition group.Pearson's correlation analysis showed that the serum MMP-13 level of ACI group was positively correlated with NIHSS(r=0.806,P < 0.01).The serum MMP-13 level of ACI group was negatively correlated with ADL(r=-0.605,P<0.01).6.The serum miR-126 level is negatively correlated with the serum MMP-13 level(r=-0.704,P<0.01).Conclusion:1.The serum miR-126 level is negatively correlated with disease severity of atherosclerosis cerebral infarction.MMP-13 level is positively correlated with it.2.The serum miR-126 level and MMP-13 level maybe a novel biomarker for predicting and diagnosing atherosclerosis cerebral infarction.
