[Objective] Gastric cancer(GC)has high incidence,low early diagnosis rate and poor prognosis.The key mechanism of GC development is still unclear.Selenium-binding protein 1(SBP1)is a GC-related antigen screened by serum proteomics analysis in our prophase research.It has been reported that SBP1 is reduced in various tumors and may be involved in the development of GC.Therefore,we will explore the expression of SBP1 in human GC tissues and serum,analyzing the relationship between SBP1 and clinicopathological features and prognosis,and construct lentiviral vector over-expressing SBP1,which provides an experimental foundation for the further study of the role and mechanism of SBP1 in the development and progression of GC.[Methods] 1.The expression of SBP1 in human GC cells(SGC-7901 and MGC-803 cells)and immortalized gastric mucosa cells(GES-1 cells)was analyzed by immunohistochemistry(IHC)and Western-blot.2.The expression of SBP1 in GC,adjacent tissues and normal gastric mucosa and the clinical significance were analyzed 3.The expression level of SBP1 protein in serum of GC patients and control group was detected by enzyme-linked immunosorbent assay(ELISA)method.4.The primers of SBP1 gene were designed for amplification of SBP1 gene fragment by polymerase chain reaction(PCR).The pCDH-CMV-EF1-copGFP lentiviralvectors were digested by Eco Râ… enzyme,which was then linked to the SBP1 gene fragment by using the ligase.The positive clones were identified by enzyme digestion and DNA sequencing.[Results]1.The expression of SBP1 protein in GC cells was significantly lower than that in immortalized gastric mucosa epithelium(P<0.05),but there was no significant difference between the two GC cells.2.The expression of SBP1 in gastric carcinoma was significantly lower than that in normal gastric mucosa and adjacent tissues(P<0.05)and its concentration was not related to gender,age and tumor size,but related to differentiation degree,lymph node metastasis and TNM stage.3.There was no significant difference in serum SBP1 level between GC patients and normal control group.The serum SBP1 levels in post-operative GC patients were significantly lower than those in pre-operative GC patients(P<0.05).4.SBP1 gene was amplified and successfully bound to the pCDH-CMV-EF1-copGFP lentiviral vectors.The sequence of recombinant plasmid was comfired correct by enzyme digestion and DNA sequencing.[Conclusions] 1.The expression of SBP1 protein in GC was decreased,its expression was related to the differentiation,lymph node metastasis and TNM stage of GC,and the reduction of SBP1 was associated with poor prognosis in GC.2.There was no significant difference in serum SBP1 level between GC patients and normal control group.The serum SBP1 levels in post-operative GC patients were significantly lower than those in pre-operative GC patients 3.The lentiviral vector over-expressing SBP1 gene was successfully constructed,which provides an experimental foundation for the function and mechanism of SBP1.
