Experimental Study Of Yishen Huoxue Recipe On Adenine Induced Renal Failure In Rats

This study by intragastric administration of high concentration of adenine induced chronic renal failure(CKD)rat model,through the observation of Yishen Huoxue Decoction and routine dose of losartan for adenine,the symptoms of CKD rats,the kidney weight and body weight ratio,serum creatinine,urea nitrogen,serum cystatin C and other laboratory indexes and histopathological observation.Study on the protective effect of adenine in CKD rats.Objective: To observe the preventive and therapeutic effects of Yishen Huoxue Recipe on renal function in rats with adenine induced renal failure,and to provide experimental data for further clinical application.Methods:(1)Laboratory animals: SPF male healthy 45 rats.(2)Model making: the body weight of rats(150±20)g,Bred in the animal laboratory of Henan Traditional Chinese Medicine Hospital of clean grade,45 rats before the experiment in the laboratory were adapted for 1 weeks,8 rats were randomly selected.The normal group,the others were the model group,the model group model group rats,CKD model the next day was using intermittent gavage adenine,rats in control group were injected with physiological saline by intragastric administration.The modeling time is 28 d,twenty-ninth days in the model group were randomly selected 5 rats were sacrificed after blood tests,creatinine,and kidney of rats,rats serum creatinine levels were higher than those of normal high limit 9.7 mmol/L,renal tissue sections under light microscope showed glomerular sclerosis,renal interstitial fibrosis of chronic renal failure,said the successful preparation of the model.(3)The drug intervention groups: the model group extremely the remaining rats were stratified according to body weight were randomly divided into 4 groups,respectively Yiqihuoxue Decoction high dose group,Yishen Huoxue Decoction and Western medicine conventional dose group(Losartan)group and model group,all rats were recorded daily at about about 16 of all rats were orally administered 1 times,a total of 8 weeks of drug intervention time.The high dose group,the conventional dose group and the Losartan group were given 49.97g/kg/d,12.49g/kg/d,and 9mg/kg/d,respectively.The model group and normal group were given normal saline to 10 m L/kg.After 8 weeks of administration,all rats were fasted for about 24 h,blood and tissue samples were collected and analyzed.Results:Compared with normal rats,the rats serum creatinine,urea,serum cystatin C values were significantly decreased(P<0.01);Yishen Huoxue Decoction high dose group rats serum creatinine,urea,serum cystatin C,kidney coefficient(left kidney weight rats and its weight ratio),decreased significantly the model group(P<0.01),drug intervention group three between the comparison of optimal Yishen Huoxue Decoction high dose effect(P<0.05);the rat renal biopsy under light microscope: Yishen Huoxue Decoction high dose of the light microscopic glomerular,infiltration of inflammatory cells is relatively small.Conclusion: Yishen Huoxue Prescription can improve the renal function of CKD rats.
