Reserach Purpose:To observe the effect of kidney-nourishing,blood-activating,phlegm-resolving,andresuscitation-inducingDecoction?KBPRD?on the the serotonin?5-HT?1B and 2C receptors in the cochlear spiral ganglion Neurons?SGN?and explore its mechanism,Then To investigate the effects of KBPRD on serum calcium?Ca?,phosphorus?P?,iron?Fe?,plasma cortisol,dopamine and cAMP Cyclic guanosine monophosphate?cGMP?,cAMP/cGMP ratio changes in serum of tinnitus rats.to provide experimental evidence for the clinical application of KBPRD for the treatment of tinnitus.Research methods:60 SPF SD male rats were selected and their body weight was 250?±50?g.The rat model of tinnitus was established by intraperitoneal injection of sodium salicylate in rats and behavioral experiments of"drinking water inhibition"Rats were randomly divided into normal group 10?saline group?,model group 50?sodium salicylate group?,Then the model group was divided into TCM group?kidney-nourishing,blood-activating,phlegm-resolving,andresuscitation-inducingDecoction,low,mediumandhighdose group?,5-carboxamide)group)10,the rats in low-,middle-,and high-dose Chinese medicine groups were given gastric administration of KBPRD at dosage of(5.5,11,22 g·kg^-1·d^-1)respectively,the rats in western medicine group were given gastric administration ofcarbamazepine at dosage of 5mg·kg^-1·d^-1),the rats in model group were given gastric administration of 2 mL of normal saline,the rats in model group and normal group were given gastric administration of 2 mL of normal saline,once every day,treatment time covering 8 weeks.by using immunohistochemistry technique to detect the express of cochlear spiral ganglion neurons of5-HT1B?2C in a rat model of tinnitus,then take the peripheral blood,calcium,phosphorus,iron were measured by automatic biochemical Analyzer,and plasma cortisol,dopamine,cAMP and cGMP were measured by enzyme-linked immunosorbent assay?Elisa Kit?.Reserach results:?1?generally:After 8 weeks of corresponding drug intervention,the body weight of Chinese medicine group,especially traditional Chinese medicine high-dose group increased significantly compared with model group,with increased food intake,bright hair and sensitive activities.The body weight of model group was significantly reduced,the spirit was sluggish,the daily activities decreased,Food intake,drinking water were reduced than the traditional Chinese medicine group.?2?Compared with the normal control group,the average noise reflex regression period of the model group was significantly reduced in the"drinking water inhibition"experiment,of which 10 were the control group,44 were successful in modeling,and the success rate was 88%.?3?After 8 weeks of the corresponding drug intervention,the expression of 5-HT1B and 2C in the SGN of the cochlea of rats after tinnitus modeling was successfully given.Compared with the normal group,the 5-HT1B in the model group decreased and the 2C increased significantly.Compared with the model group,the expressions of 5-HT1B in SGN of cochlear SGX group and2C group were significantly decreased in western medicine group and traditional Chinese medicine group.?4?After 8 weeks of the corresponding drug intervention,Compared with the model group,Sodium salicylate group decreased serum calcium,iron,and phosphorus concentrations increased.And Plasma cortisol,dopamine levels dropped,cAMP decreased cGMP content increased cAMP/cGMP ratio decreases?all P<0.05?.Compared with the model group,the KBPRD improves the tinnitus in rat serum calcium,iron,and contents of plasma cortisol,dopamine and cAMP?cAMP/cGMP content and reducing serum phosphorus and cGMP?all P<0.05?.Reserach Conclusion:Tinnitus rat cochlea SGN 5-HT1B decreased,2C expression increased,The KBPRD has effects on tinnitus of rats mechanism being correlated with the reduced expression of 5-HT1B and increased the expression of 5-HT2C in SGN of tinnitus rats.And the same time,The KBPRD improves the tinnitus in rat serum calcium,iron,and contents of plasma cortisol,dopamine and cAMP?cAMP/cGMP and reducing serum phosphorus and cGMP may be improved tinnitus Hematology-related evidence.
