Objective:The microRNAs are a non-coding small molecule RNA about 22 to 28 nucleotides which play an important role in the post-transcriptional level and translational level of gene regulation.MiR-21 has been shown to participate in many physiological and pathological regulation.Hepatic oval cells(HOC)is the original and liver pluripotent stem cells,mainly in the liver bile ducts or between Hering tube that terminal bile duct area.Hepatic oval cells with strong self-replication,clonal proliferation and differentiation in vitro,when severe liver damage or mature liver cell proliferation was inhibited liver regeneration is mainly done by oval cell proliferation and differentiation.The purpose of this study is to explore the role and mechanism of miR-21 in the EMT of the rat hepatic oval cells.Methods:The expression of miR-21 was enhanced and inhibited by transfection of rat hepatic oval cells with lentivirus.The transfection efficiency was observed by fluorescence microscopy and the expression of miR-21 was detected by PCR(empty vector group,miR-21 enhanced group,miR-21 inhibitory group).Extraction of proteins in transfected hepatic oval cells.The expression of Smad7 protein was detected by Western blot,and the expression of Smad7 mRNA was detected by PCR.The proliferation of three groups of hepatic oval cells was detected by cck-8 method.Results:Lentivirus was transfected into the hepatic oval cells and the stable cell lines were screened.The miR-21 expression by PCR was enhanced in the miR-21-enhanced group,and the miR-21 expression was inhibited by miR-21-inhibited group.Western blotting showed that the expression of Smad7 protein was decreased in miR-21-enhanced group,and the expression of Smad7 protein was decreased in miR-21 inhibited group.PCR results showed that the trend of Smad7 mRNA and miR-21 expression trend consistent.Conclusion:Overexpression of miR-21 promotes the process of hepatic oval cell proliferation through Smad7.
