Objective: To detective the theory of psoralen anti postmenopausal osteoporosis from the OPG/RANKL/cytokine network regulation on astration of female rat.Methods: using 72 three month old female SD rats to established the postmenopausal osteoporosis model,randomly divided 12 rates into sham operation group,then the 60 rates randomly divided into model group, valeric acid estradiol positive control group, the zhuangguzhitong prescription media concentration group, psoralen high dose group, psoralen low dose group and each group have 12 rates,Then intragastric administration after one week later, the intragastric administration of drugs of psoralen high, low dose group were administered 8mg/kg, 4mg/kg; the estradiol valerate positive control group is 0.021mg/kg of estradiol valerate, zhuanggu zhitong prescription group is 6.6mg/kg,and the sham operation group and the model group are intragastric administration distilled water for 1ml/100 g, ntragastric administration for 13 weeks.Then every group randomly chose 6 female rats to extract the mRNA sample of femur tissue and start to get RT-PCR test the expression of mRNA of IL-10 IL-17 TNF-? TGF-? ER?and 6 female rats to collect blood from abdominal aorta and centrifuge(500g) after solid and get the serum to use Elisa to test the concentration of OPG RANKL IL-10 IL-17 TNF-? TGF-?and E2.The OPG and RANKL which usded the femur of RT-PCR in the other side were detected by ELISA.Results: RT-PCR tests the expression of mRNA of femur tissue, compared with the model group, groups of IL-17 mRNA expression were statistically significant;IL-10 mRNA,TNF-? mRNA,TGF-?mRNA and ER?mRNA expression except the psoralen low dose group have statistical significance. ELISA enzyme-linked immunosorbent adsorption assay and compared with the model group, groups of OPG, RANKL, IL-10, IL-17 and TNF-? concentration has statistical significance, E2, TGF-? in addition to psoralen low dose group were statistically significant.Conclusion: 1.Psoralen by regulating estrogen and estrogen receptor levels play a role in anti postmenopausal osteoporosis;2.Fructus Psoraleae can increase the content of ovariectomized rats serum and bone OPG, decrease the content of RANKL;3.Psoralen by affecting the OPG/RANKL/ cytokines play against postmenopausal osteoporosis.
