Preliminary Study On The Chemical Constituents And Anti-inflammatory Activities Of Ethyl Acetate In Lungs

Feijingcao is the dry whole grass from Saniculalamelligera Hance,which has effect of expelling wind,expectorant cough and regulate the menstrual function.It widely distributes in Sichuan and the southwest of China.Feijingcao is a medicine for treatment of the cold,cough,asthma,irregular menstruation,amenorrhea,dysmenorrhea,sores,traumatic swelling,bleeding etc.The preliminary study found that Feijingcao had good effect of relieving cough and eliminating phlegm.In order to find the material basis of anti-inflammatory effects,the isolation and structure identification of Feijingcao ethylacetateextract has been completed,fourteen compounds from the plant were identified,including two new compounds andtwelve known compounds by using normal,reverse phase silica gel,macroporous resin,MCI,Sephadex LH-20,TLC preparation and HPLC preparation.They are saniculamins A(1),saniculamins B(2),derivatives brosimacutin(3),6-flavonol(4),eucomol(5),Isopsoralen(6),Isopropyl aldehyde(7),12-hydroxybakuchio(8),22-angeloyl-R1-barrigenol(9),5,6,7,8,4’-Pentamethoxyflavone(10),3,5,6,7,8,3,4’-heptamethphoxyflavone(11),isobavachin(12),isoquiritigenin(13),Furan(2,3,7,6)-4’-hydroxy dihydrogen flavonoids(14)After the separation of feijingcao ethyl acetate,combined with previous reports,we tested 2 new compounds(compound 1,2),and one of the reported compounds(compound 3),inhibition of nitric oxide release from ratsmacrophages of RAW264.7 induced by lipopolysaccharide.Results showed that compound 1 and compound 2 had a good effect on inhibited nitric oxide released,they had a certain anti-inflammatory activity,revealed the material basis for anti-inflammatory feijingcao ethyl acetate.This research provided a scientific basis for the study of anti-inflammatory activity and benified development of feijingcao resources.
