| Objective:To observe the effect of the behavior,gastric emptying,gastrin,somatostatin and 5-HT of the depression model rats,which are intervened by Electroacupuncture on Zusanli etc.To explore the mechanism of depression and gastric motility disorders intervened by acupuncture based on dispersing stagnated liver qi for regulating stomachMethod:A total of 60 healthy rats were randomly selected as the blank group,and the remaining 44 rats were used to make the model.Modeling methods:using CUMS combined with single cage feeding.After the modeling,based on Body weight,food intake,open field test,gastric electrical test,12 mice that did not meet the requirements were removed.8 samples were randomly selected from the blank group and the model group,through the observation of gastric emptying of these 16 rats,to determine the success of the model whether or not.The remaining 24 rats were randomly divided into model group,acupuncture group and drug group.Intervention method:blank group without any treatment;model group were bound only;The acupuncture group was treated with electro acupuncture,such as gan shu,once a day for 14 days;The drug group with Prozac suspension gavage,1 times/d,administered continuously for 14 days.After the end of the experiment in the hippocampus of rats was detected by immunohistochemical method,to observe the morphological changes of hippocampus were observed,immunohistochemical method of 5 hydroxytryptamine(5-HT)levels;abdominal aortic blood after centrifugation,ELISA double antibody sandwich enzyme(ELISA)detected in rat serum GAS,SS content.Results:1 The model rats show listlessness,decreased activity,hair dry,loose stools.Food intake and body weight were significantly lower than those in the blank group (P<0.01).Delayed gastric emptying rate compared with the blank group(P<0.05),open field showed that the horizontal displacement of the model group was lower than that of the blank group(P<0.01).The number of vertical direction perpendicular to the model group decreased significantly,and lower than that of the control group(P<0.01);gastric electrical detection indicated that the model group compared with blank group amplitude decreased obviously(P<0.05),percentage of normal slow wave decreased significantly(P<0.01),gastric bradycardia percentage increased significantly(P<0.01),no gastric electric rhythm percentage increased significantly(P<0.01),suggesting that the model was successful.2 Observe the morphological changes in hippocampus of rats:control group cell morphology,cell membrane and nucleus of clear boundaries,and in the model group,the visible part of irregular cell morphology,the nucleus cytoplasm is not clear,the number is less.In the acupuncture group,irregular cell number less than the model group that stain showed similar with blank group.The drug group,irregular cell number decreased compared with model group,staining showed similar with blank group.3 5-HT in the hippocampus of detection:compared with the control group,the low content of the model group,the difference was statistically significant(P<0.05),acupuncture group,drug group lower than control group,but no statistical significance(P>0.05).Compared with the model group,acupuncture group,drug group was significantly higher,the difference was statistically significant(P<0.05),and the acupuncture group,drug group were higher obviously,but the difference was not statistically significant(P>0.05).4 SS of rats in serum:compared with the blank group,model group,low drug group content of SS(P<0.05);acupuncture group was lower than control group,but no statistical significance((P>0.05).Compared with the model group,the acupuncture group was significantly higher(P<0.05);drug group compared with the model group,but no statistical significance.(P>0.05)and acupuncture group than in the drug group was lower(P<0.05).5 GAS of rats in serum:from control group,model group,acupuncture group,drug group GAS was lower(P<0.01).Compared with the model group,the drug group was significantly lower(P<0.01);acupuncture group compared with model group content was significantly higher(P<0.01).6 The gastric emptying of rats:compared with control group,model group,drug group and the gastric emptying rate was significantly lower(P<0.05),gastric emptying in acupuncture group than the control group the rate is low,but the difference was not statistically significant(P>0.05).Compared with the model group,drug group,gastric emptying rate is low,but no statistical significant differences(P>0.05),acupuncture group compared with model group,the gastric emptying rate(P<0.05).And the acupuncture group,drug group was significantly lower,the difference was statistically significant(P<0.05).7 Open field test results as follows:2 weeks after treatment,and the control group,the total horizontal displacement model group activity is significantly lower(P<0.01).Compared with the model group,acupuncture group and drug group level movement distance increased significantly(P<0.01);acupuncture group was lower than that in the control group,but no statistically significant difference(P>0.05).(2)in the 2 week after the treatment,and the control group,drug group,model group,acupuncture group,the number of vertical vertical direction is significantly lower(P<0.01).Compared with the model group,drug group,acupuncture group,the number of vertical high vertical direction(P<0.01)compared with the drug group.Upright,perpendicular to the direction of the low frequency acupuncture group,but the difference was not statistically significant(P>0.05).Conclusion:1 Electro-acupuncture can improve the depressive symptoms and gastrointestinal motility disorder symptoms of depression model rats.2 Electro-acupuncture can increase the positive expression of 5-HT in hippocampus of model rats.It may be the mechanism of the improvement of depressive symptoms in depression model rats.3 Electro-acupuncture can increase the content of SS and GAS in serum of model rats.It may be the mechanism of improving the gastrointestinal motility disorder in the rat model of depression. |
