| The theory of yin and yang in Huangdi Neijing emphasizes the spirit of yang on the basis of a great deal of yin and yang relationships and the deficiency of Yang Qi is an important cause of human disease.Yang-deficiency constitution is closely related to the occurrence of many diseases in clinic.Kidney yang is the root of Yang,if the kidney yang is deficient,then the body Yang Qi is deficient.Therefore,warmly invigorating kidney yang can play a very good role in improving Yang-deficiency constitution and preventing diseases occurrence.Due to the slow improvement of Yang-deficiency constitution,we need to explore an effective and safe method to correct the Yang-deficiency constitution,Jingui Shenqi pill(JGSQP)a classic prescription for treatment of kidney yang deficiency,this experiment is to compare the pharmacodynamics of two dosage form of Jingui Shenqi pill(JGSQP)and its pharmacodynamic mechanism.1 ObjectiveFor a wide range of Yang Deficiency Syndrome(Yang-deficiency constitution),uesing the Paste and Decoction of Jingui Shenqi pill(JGSQP)treating kidney yang deficiency rats made through adenine by perfusion in order to compare the effect of two kinds of dosage forms on kidney yang deficiency rats and its pharmacodynamic mechanism.2MethodsForty male SD rats of clean grade were selected,the rats model of kidney yang deficiency were induced by intragastric administration of adenine 200mg/kg and divided into normal control group,model control group,paste intervention group and decoction intervention group.In the paste intervention group,the paste which was boiled and dissolved to a suitable temperaturewas given by intragastric at 18.36g/(kg·d);in the decoction intervention group,the decoction was given by intragastric at 18.36g/(kg·d);in the model control group,distilled water was given at 10mL/(kg·d);in the normal control group,rats were fed normaly.The intervention was lasting for 6 weeks.The appearance of rats were observed and weight,urine output,sex hormones e.g.Serum testosterone(T),estradiol(E2),luteinizing hormone(LH),human chorionic gonadotropin(HCG),adrenal cortex hormones e.g.adrenocorticotropic hormone(ACTH),cortisol(CRE),17-hydroxycorticosteroid(17-OH-CS),thyroid hormones e.g.three triiodothyronine(T3),four thyroid ammonia(T4),and other hormone index sperm quality were tested and the kidneys were stained with HE,Masson and electron microscope to observe the pathological changes of the kidneys.In the end,curative effect of two dosage form of Jingui Shenqi pill(JGSQP)was evaluated.3 Results3.1 General conditionThe model group showed the symptoms of kidney yang deficiency,such as wight loss,slow reaction,decreased activity,arched curled up,chills,sparse body hair loses luster.After the treatment of the paste and the decoction,the intervention was obviously improved.We can see increased activity,the phenomenon of curling up disappeared,body hair gradually restore luster.3.2 Renal morphology3.2.1 light microscopeThe rats in the model control group showed the following changes:(1)glomerular abnormalities:decreased glomerular number,glomerular atrophy,enlargement of renal capsule,glomerular capillary lobes and atrophy,inreaded extraglomerular mesangial cell.(2)renal tubular acidosis:renal tubule number decreased,deposition of 2,8-two hydroxy adenine in the renal tubule especially in proximal tubule,enlarged tubular lumen with crystalline deposits,epithelial thinner,a small amount of exfoliated epithelium was seen in the lumen.(3)renal interstitial lesions:fibrous tissue hyperplasia in renal interstitium,visible foreign body granuloma and fibrosis lesion formation,severe fibrosis of renal tissue?The light microscope was obviously improved in the paste group and the decoction group.We can see:(1)glomeruli:a slight reduction in the glomerulus,a mild atrophy of the glomerulus,a normal renal capsule,and no atrophy of the glomerular capillaries.(2)renal tubules:the number of renal tubules was normal,and there were no obvious crystals in renal tubules.(3)renal interstitium:mild fibrous hyperplasia in the renal interstitium.3.2.2Electron microscopyCompared with the normal group,the model group showed the following changes:(1)glomerulus:The necrosis of endothelial cells,the collapse of the lumen,the fusion of the foot processes and microvilli,basement membrane thickening,capillary collapse.(2)Renal tubule:The microvilli of proximal convoluted tubules were sparse and disordered,Partial epithelial cell Small vacuolar degeneration,mitochondrion vacuolar degeneration necrosis,white crystals were found in the cytoplasm of epithelial cells and in the lumen of proximal convoluted tubules.Paste group showed:partial foot processes in glomeruli fused,part of the foot processes was interspersed with each other into a fence like,clinging to the capillary basement membrane.Mild thickening of basement membrane could be seen.The density of microvilli in renal tubules was normal and a small amount of white crystals were found in the proximal tubules.Decoction group:small part of the glomerular process restored the palisade structure,the majority of the foot processes were fused,the basement membrane was thickened,the microvilli of the proximal convoluted tubules were still sparse,and the white crystals were found in the lumen of the proximal convoluted tubules.3.3 Hypothalamus-pituitary-target axis3.3.1 Hypothalamus-pituitary-gonadal axisCompared with the normal group,the content of LH in model group was significantly decreased(p<0.05),LH was significantly increased in the decoction group and paste group(p<0.05),Compared with the decoction group,the LH value of the ointment group was higher.Compared with the normal group,the content of E2 in model group was significantly decreased(p<0.05),compared with the model group,the E2 of decoction group increased significantly(p<0.05),and the content of paste group also increased,but there was no significant difference.Compared with the model group,the content of T increased significantly(p<0.05)in the decoction group and paste group.Compared with the normal group,the content of 17-OH-CS in model group was significantly higher(p<0.05).After drug intervention,the decoction group was significantly lower than that of the model group(p<0.05),and the proportion of the paste group was decreased,but there was no significant difference.3.3.2 Hypothalamus-pituitary-thyroid axisCompared with the normal group,the content of T4 in model group was significantly decreased(P<0.05).Compared with the model group,the content of the paste group was significantly increased(P<0.05),and there was an increasing trend in the decoction group,but there was no significant difference.3.4 Sperm qualityThe sperm motility rate of model group was significantly lower than that of normal group(p<0.05),and that of the paste group was significantly higher than that of model group(p<0.05).Compared with the normal group,the level of sperm in the model group was significantly decreased(p<0.05),which was significantly higher(p<0.05)in the paste group,and there was an increasing trend in the decoction group,but there was no significant difference.4Conclusion1 Jingui Shenqi pill decoction and paste can obviously improve the rats of kidney yang deficiency whole life signs,including its arched curled up,sparse body hair and less activity and other typical kidney yang deficiency syndrome,improve the morphological and pathological damage of rat kidney induced by adenine and the function of hypothalamus pituitary gonad axis and hypothalamus pituitary thyroid axis were improved,so Jingui Shenqi pill can significantly improve kidney yang deficiency syndrome.2 In comparison,Jingui Shenqi pill paste is better than Jingui Shenqi pill decoction in improving the content of LH and T4 and the sperm motility of the kidney yang deficiency model.But,Jingui Shenqi pill decoction is better than ingui Shenqi pill paste in enhancing E2 content. |
