| Objective:1.Based on TCMIAP(inheritance auxiliary platform)V2.5,to find the rule of the prescriptions of the cerebral apoplexy aura,according the data mining.The contents of the mining include syndromes analysis,four-qi analysis,five-wei analysis,drug frequency analysis,commonly used drug analysis,Core combination and the analysis of new combination.Through the data mining,we can get to know more about the TCM treatment on cerebral apoplexy aura.2.To investigate the change of expression of IRAK-2mRNA and the apoptosis in the kupffer cell in hepatic stress injury of acute cerebral hemorrhage.Methods:1.The data mining is aim to collect the TCM prescriptions of the cerebral apoplexy aura,which are publised in recent 20 years.The data source is from the CNKI 1997-2016 TCM herb treatment of the cerebral apoplexy aura.(TIA/Cerebral Ischemia).After the abnormal data cleaning,we get 141 documents and 119 prescriptions.To ensure the accuracy of the input of the prescriptions,we take steps to double-check the data.2.The model of acute cerebral hemorrhage was established.The animals were randomly put into 3groups:Medical-treatment-group:rhubrab,brave ry,south-star,stone-calamus,cattail pollen,notoginseng,leech,cerebral hemorrhage group,blan k control group.And each group was further divided into 4 time-point sub-groups of a fter modeling for 6?12?24?48hours.The expression of IRAK-2mRNA in the kupffer cell was measured,the hepatic content of tumor necrosis factor-alpha(TNF-?)and the ap optosis were detected.Result:1.The result of the data mining shows the qi-deficiency and blood stasis?the liver wind agitation?yin-deficiency of the liver and the kindey?the phlegm-dampness blocking collaterals are the fundamental pathogenesis of cerebral apoplexy aura.Medications mainly are made up with the warm and cold drugs;the most of the frequency in five flavors is the sweet taste of drugs,by the liver channel mainly,heart and spleen and kidney followed;Chuanxiong,Astragalus,Poria,red peony are much more used to invigorate the circulation of blood,nourish Qi,enrich the blood;obtained 22 groups high-frequency drug combinations,according to the association rules analysis,obtained 11 rules of medication;clustering algorithm on the entropy hierarchical to extract 18 core combinations,9 new prescriptions.2.Compared with the blank group,the expression of IRAK-2mRNA in the kupffer cell in the cerebral hemorrhage group at after modeling for12h,24h,48h time-points were reduced markedly(P<0.01).Group Treatment increased markedly in 3 time-points,after modeling for 12h,24h,48h time-points,compared with the cerebral hemorrhage group(P<0.01).The expression of IRAK-2mRNA in the kupffer cell was negatively correlated with the changes of hepatic content of TNF-a.Compared the content of TNF-aof medicine treatment group with the cerebral hemorrhage model group on four time points,the content were reduced,and were statistically significant(P<0.01);from the scart plot,the quantity of early-apoptosis cells in the model group is more than the control group in 12 hours,24 hours.The number of apoptotic cells in the treatment group was lower than that in the model group at 12 hours and 24 hours.There was significant difference in 95%confidence interval between the groups at each time point(P<0.05);Conclusion:1.Based on the results obtained from data mining,it shows that the pathogenes of the stroke precursor is deficiency of Ben and repletion of Biao.The empty of Qi and yin of the liver and kidney is reason,blood stasis and phlegm is outer phenomenon.The syndrome distribution,Qi-deficiency and blood stasis?the liver wind agitation?yin-deficiency of the liver and the kindey?the phlegm-dampness blocking collaterals always living in the first onset of stroke.From the frequency analysis,the herbs which nourish qi and invigorate the circulation of blood give priority to use.At the same time,we can clean the retention of wind and phlegm in the liver channel.According to the association rule of Chinese medicine inheritance platform,11 rules and 18 core combinations were obtained,and 9 new prescriptions were given theoretical guidance from the different angles to the treatment of the cerebral apoplexy aura.However,the theoretical guidance of traditional Chinese medicine is inseparable from the test of the efficacy of practice.This study lacks of the combination of theory and practice,which is the most important target to master the ability to take participate in clinical practice,summarize the experience.The aura of the stroke is often overlooked by a transient performance.Our community need to take more steps to promote the prevention and treatment of the stroke aura related education.2.The increase the expression of IRAK-2mRNA may plays a significant role in the inhibition of endotoxic liver injury,and Ditantongyu measure has some protective effect on kupffer cell. |
