The Effects Of Acupuncture On The Contents Of GAS, SS And MT In Stress Ulcer Model Rats Were Investigated From The Perspective Of Brain-intestinal Axis

Stress Ulcer(SU)is causing by psychological stress,extensive surgical procedure,severe injury or other critical disease is characterized by acute erosive gastritis or ulceration of stomach,esophagus and duodenum mucous membrane.It is also named as stress-related mucosal disease(SRMD),in TCM which is classifying as "epigastric pain","gastric upset"and "acid regurgitation" area.As the well developing of the society,the pressures come from working and living keep raising.Therefore,SU become a common disease in today's clinic.The occurrence of gastrointestinal disease is not only reflect at the digestive system,but also accompany with somnipathy.A large number of ancient TCM document was recorded the correlation between gastrointestinal disease and sleep."Yellow Emperor"proposes "Stomach disharmony results in disturbed sleep".In the TCM philosophy,spleen is the origin of the qi-blood and the foundation of acquired constitution.Insomnia can lead by dysfunction of qi in transformation causing by spleen and stomach physical weakness,pyrophlegm due to deficiency of spleen while affect the state of mind and food stagnation results in stomach qi upward floating.Morden medicine explore some interrelation of gastrointestinal and sleep from the mechanism of brain-intestinal axis and brain-intestinal interaction.This theory has been proved by clinical researches.Liu YaNan and other researchers found a positive result when adopting the conbination acupuncture treatment of tranquilizing by nourishing the heart and strengthening the spleen and stomach for insomnia.However,lack of studies on the pathogenesis of how the stomach affect the quality of sleeping,the relationship between GAS,SS and MT as the brain-gut peptide.The mechanism of acupuncture regulation of gastrointestinal disease and sleeping still not clear.Therefore,this experiment adopted restraint water-immersion stress(RWIS)to copy SU model rats.Using the acupuncture treatment to observe the relationship among SU and sleep and to study the mechanism of acupuncture therapy.ObjectiveTo investigate the acupoints of(ST36),(CV12)to the restraint water-immersion stress copied SU model and alter containment of GAS,SS and MT in peripheral blood.And for studying the correlation of this three factor and the mechanism of acupuncture therapy.Using brain-intestinal axis to analysis the impact of gastrointestinal disease toward the quality of sleeping and the dependence among them,for the theory "Stomach disharmony results in disturbed sleep" proving modern scientific evidence.MethodsThe study was performs after 28 adult health male SD rats were allowed to acclimate for 1week.They were randomly divided into normal group,model group,acupuncture treatment group,drug group by random number table(n=7),adopting the restraint water-immersion stress(RWIS)to make the gastric ulcer model.For rats in acupuncture treatment group,acupoint at "Zhongwan"(CV12)and "Zusanli"(ST36)were chosen for needling 20 minutes,manipulating 30 seconds for each 5 minutes.The rats in drug group were tread with omeprazole enteric-coated tablets(0.2 mg/kg)by oral administration once for each of a day.After 5-day treatment,the morphology of gastric mucosa in rats were observed.After the last treatment,a sleep model was established by intraperitoneal injection of sodium pentobarbital(40 mg/kg).The Guth Scoring method was adopted to measure the gastric ulcer area of the rats,and the rats' gastric ulcer index(ulcer index,UI)was calculated.The contents of GAS and MT in the serum and hypothalamus were detected by ELISA.ResultsSleep duration:Compare with the normal group,the sleeping duration of model group were significantly much lower(p<0.05).The duration between model group and the drug group without significantly comparison.Compare with the drug group,the sleeping duration of acupuncture treatment group were significantly much higher(p<0.05).Gastric ulcer index score:The gastric mucosal injury index of the model group was significantly higher than that of the normal group(P<0.05),showing the successfully modeling.Compared with the model group,the gastric mucosa hyperemia and the injury index were significantly decreased in the acupuncture group and the drug group(P<0.05).The index between drug group and the acupuncture treatment group without significantly comparison.Mucosal morphology:Blank gastric mucosal surface is smooth,no congestion;model gastric mucosa darker,showing a lot of spotting lesions,mucosal folds shallow surface defects in the formation of ulcers;bleeding gastric mucosa acupuncture group less than the model group,smaller ulcers,mucosal folds still clear,mild defect,some near normal.Serum,containment of GAS,SS and MT at hypothalamus:Compared with the normal group,the levels of GAS in the serum and hypothalamus of the model group were significantly increased(P<0.01);Compared with the model group,the levels of GAS in serum and hypothalamus of acupuncture group and drug group were significantly decreased(P<0.01).The levels of MT in serum and hypothalamus were significantly higher in normal control group.Compared with the model group,the expression of MT and SS in serum and hypothalamus of acupuncture group was significantly increased(P<0.01).Conclusion(1)The restraint-immersion water(RWIS)stress method can cause gastric mucosal damage in rats,the formation of stress ulcers,which can affect sleep in rats and reduce the duration of sleep in rats.Sleep time is negatively related to the degree of gastric mucosal injury.(2)Needling the acupoints of(ST36),(CV12)can reduce peripheral and central gastrin content,increase somatostatin content,reduce gastric ulcer injury index,reduce ulcer area,reduce mucosal hyperemia and edema,and can effectively protect gastric mucosa and treat stress ulcers.(3)Acupuncture can improve sleep disorders in rats caused by stress ulcers.(4)Acupuncture is superior to the drug treatment group in treating sleep disorders caused by gastric ulcers and gastric ulcers.Gastrointestinal disease and sleep were closely related.Needling the acupoints of(ST36),(CV12)could treat the gastrointestinal disease,inhibit the stimulation of gastric acid which showing the positive effects on reducing the injury of mucous membrane.Meanwhile,utilising "Harmony Stomach" for reaching the aims of "relieve uneasiness of mind and body tranquilization" to regulate the sleeping quality.The mechanism of acupuncture therapy might be reduced the central and periphery GAS and raised the continence of SS and MT at central and periphery through Neuro Endocrine Network.Therefore,regulating the brain-intestinal axis could meet the dynamic equilibrium of Neuro Endocrine system.Increasing the blood fluid of stomach mucous membrane,inhibiting the stimulation of gastric acid,improving the recovery of the ulcer,anti-stress effect to keep down the concentration of cortisone during stress could improve the sleeping quality.
