| Objective:Though the acupuncture on the Shu point,Yuan-primary point of rats' liver and kidney as well as the Mu point of their large&small intestine respectively,to observe these three points'effects on the concentration change of serum uric acid-SUA,blood urea nitrogen-BUN,aspartate aminotransferase-AST and alanine transminase-ALT in the Hyperuricemia(HUA)Model Rats body,and also,to observe the effect on the organizational structure of liver cells,further,to investigate the intervening effect of different points on HUA,finally,to provide scientific evidence for point selection and point combination of the clinical acupuncture for HUA.Methods:The experiment lasts for 90 days.Firstly,dividing the 50 healthy adult male Wistar rats into 5 groups at random as blank group,model group,Shu point group,Yuan-primary point group and Mu point group,and each group has 10 rats.The Hyperuricemia(HUA)Model Rats were prepared by intragastric administration,and the model making agent is the mixture of adenine(100mg/kg)and Ethamethylamine ethanol(250mg/kg).Then except the blank group,all the 4 groups started to make the model at the same time from the first day of the experiment.At the first 21 days,the rats were given the agent once a day.And 21 days after,the agent was given to the rats every two days instead until the end o f the experiment.Besides the agent given,the rats in the experiment groups:Shu point group,Yuan-primary point group or Mu point group were given acupuncture treatment at the same time.The acupuncture treatment was given every two days with the time lasted 15 minutes every time.Every 10 times of acupuncture treatment was regards as a course of treatment,and with the end of a course,the acupuncture would be suspended for 10 days,then the acupuncture would continue.Based on this cycle,there were 3 treatment courses till the end of the experiment.For the detection of indicators,the experiment detected the SUA,BUN,ALT,AST by the fully automatic biological analyzer,and used the ordinary optical microscope to observe the organizational structure of rats' liver cell,which were made into parafin section and stained by the hematoxylin-eosin(hematoxylin-eosin staining,HE staining).Results:Compared with the blank group,the BUN,SUA concentration in model group is obviously higher with significant difference(p<0.05).Observing the liver section under the electron microscope,the liver cell edema of the central veins of hepatic lobules,the cell loosen,the cytoplasm opacitas and the unclear radial arrangement of the hepatic cell cord arrangement was seen.Compared with the model group,the SUA,BUN concentration in Shu point group,Yuan-primary point group and or the Mu point group decreased at different degree(p<0.05).Observing the HE stained liver section under the electron microscope,we saw the following changes compared to those in the model groups,that was liver cell edema range of the central veins of hepatic lobules was obviously reduced,the cytoplasm opacitas improved at varying degree,there was some lymphocytes infiltrated and the radial arrangement of the hepatic cell cord arrangement was clear.Compared the 3 experiment groups of acupuncture,we got that the SUN and BUN concentration decreased largest in Mu point group(p<0.01),as for the concentration reduction of SUA,the intervention effect in Yuan-primary point group was better than that in Shu point group,however,in terms of the BUN concentration reduction,the intervention effect in Shu point group was better than that in Yuan-primary point group.Besides,both the ALT and the AST concentration was no obvious difference among the 5 groups.Conclusion:According to the experiment,we could conclusion that there is a certain influence on SUA and BUN concentration of HUA by acupuncture on Shu point and Yuan-primary point of liver,spleen or kidney and Mu point of large and small intestine.And the detailed effect is as following:1)Both the SUN and BUN concentration could reduce by acupuncture on Shu point group,Mu point group or Yuan-primary point group.2)The concentration reduction effect of SUA in groups is Mu point group>Yuan-primary point group>Shu point group.3)The concentration reduction effect of BUN in groups is Mu point group,Shu point group>Yuan-primary group.4)Both the AST and the ALT are not influenced at the HUA state. |
