| As one of the common Chinese medical herbs,Plantain seed can be used alone or combined with other Chinese medical herb,as well as taking oral and external,played an important role in the Chinese herbal medicine.The price of Plantain seed is very cheap,and Plantain seed have many functions,such as clearing heat,diuresis,eliminating phlegm,cooling blood and detoxification.At present,there are many researches about the chemical composition,pharmacological,extraction technology and detection methods of Plantain seed,but mainly concentrated on the Semen plantaginis polysaccharides and flavonoids,the reports about extraction and separation phenylethanoid glycosides are relatively few.If got high purity and high content phenylethanoid glycosides from the Plantain seed,it would be more economical and practical.This article studied the optimizing production methods of phenyethanoid glycosides,designed to explore a route which was more suitable for the industrial production.By response surface analysis,The optimum ethanol extraction conditions were found to be:optimized extraction temperature 90 ℃,ethanol concentration 70%and ratio of solvent to material 46.Under these conditions,the mean extraction yield of acteoside was 0.99%,which was in good agreement with the predicted model value.The optimum utrasonic extraction conditions ware found to be:optimized extraction temperature 58 ℃,extraction time 34 min and ratio of solvent to material 26.Under these conditions,the mean extraction yield of acteoside was 0.74%,which was also in good agreement with the predicted model value.Further,purificated the Plantain Seed extract to high purity phenylethanoid glycosides,by using of macroporous resin and preparative liquid chromatography.The optimization process was select HPD400A macroporous adsorption resin,sample amount 500 mg,the adsorption flow rate 2.7 mL/min desorption flow rate 3.5 mL/min,desorbent was 400 mL of 30%ethanol solution.After extraction and purification,obtained phenylethanoid glycosides 3.07 g,whose purity was 80%,the yield was 0.61%.In addition,researched the effects of phenylethanoid glycosides and Plantain seed extract to prevent hyperuricemia in rats by testing the SUA,SCr,BUN and XOD,thus proved that phenylethanoid glycosides had food and pharmacological value and provided a scientific basis of applicating to industrial manufacture.At last,this topic designed a production-line that annual of phenylethanoid glycosides were 100 kg,in accordance with the general requirements of food factory,designed production programs and processes,obtained the annual material cost of production was 5,73 million yuan,the annual water,power,and steam production cost was 420,200 yuan,the annual wages,benefits,insurance and manufacturing cost were 60,000 yuan,the total cost of product was 6,79 million yuan,the tax was 1,06 million yuan,the annual sales revenue was 10 million yuan,the revenue of by-product was 600,000 yuan,the profit was 2,75 million yuan;selected DN65 water pipes and DN65 steam pipelines. |
