| Cistanche tubulosa(Schrenk)Wight,which belongs to Orobanchaceae Family parasitic herb,was specified in 2005 Chinese Pharmacopoeia. It has some functions such as improving immunity, enhancing physical fatigue, improving memory, antioxidant, anti-radiation, anti-atherosclerosis and other pharmacological effects.This article is based on furtherly developing Cistanche tubulosa,which is the characteristics of resources in Xinjiang .with its main active ingredient PeG as the index components, This article study on extraction, purification, study the pharmacological effects of Cistanche tubulosa extracts on improving the ability of learning and memory in mice and the fingerprint of Cistanche tubulosa.This article provide theoretical basis for improving the processing preparation of Cistanche tubulosa’s extract, establishing quality standards and developing the functional foods.1. MethodAdopt the orthogonal test to optimize the extraction process of the PeG from Cistanche tubulosa.The extracts substances of Cistanche tubulosa were purified by macroporous colophony.study refined PeG concentrate’s pharmacological action on learning and memory in mice; use the mehhod of HPLC to establish the fingerprint of Cistanche tubulosa.2. ResultThe extraction process of the PeG from Cistanche tubulosa: liquid ratio 1:12,70% ethanol extract 2 times, each time 1 hour. the purification process of the PeG from Cistanche tubulosa: the sample concentration 4mg/ml, adsorption flow rate 1ml/min, elution flow rate 1ml/min, high diameter ratio of 1:15. The fingerprint chromatographic conditions of Cistanche tubulosa: column: Waters xterra RP18 (250mm×4.6mm , 5μm); mobile phase: acetonitrile-0.2% phosphoric acid gradient mixture (gradient elution Table 3-1); column temperature: 25℃. Experimental results show the PeG extract Cistanche tubulosa has a good capacity to improve learning and memory in mice,it can be used as deep processing of raw materials for functional products .3. ConclusionThe Content of 11.2% Echinacoside and 28.7% PeG can reach by the best extraction process. Purified by macroporous resin, Echinacoside content can reaceh 33.4%, PeG content reach 75.8%. The experiments have been shown, the extraction and purification of Cistanche tubulosa are reasonable, stable and suitable for applying to actual production.Thesis provide theoretical basise for improving the PeG processing preparation of Cistanche, developing the functional product and industrial production. The establishment of fingerprint of Cistanche Cistanche can control inherent qualityand provide basis for more scientific quality standards. |
