| Hovenia dulcis is widespread with high yield,it also grow fast with economic value.On the other hand,amino acids are essential to the human body,non-protein amino acid has a strong medicinal value.In order to make better use of Hovenia dulcis,this paper researches on free amino acids of the Hovenia dulcis which has not been explored much,we extract and isolate free amino acids from the fleshy fruit stalk of Hovenia dulcis,and testify its beneficial function,the main research contents are as follows:1 Using Hovenia’s fleshy fruit stalk as raw material,single factor experiment were selected for the extraction temperature,extraction time and liquid ratio and extraction times,with comprehensive consideration of other influencing factors in the experiment,we find out the key data for the response surface experimental design method through the comparison of free amino acid yield.Among them,extraction times,extraction time and the ratio of liquid to solid center points were:two times,60 min and 20 ml/g,extraction temperature 100 ℃.2 Base on single factor experiment,we choose extraction times,extraction period and the ratio of liquid to solid as three parameters to do 3 factors 3 levels CCD experiment.Picking amino acid yield as response value:we find out the perfect combination for our purpose are as follows which could provide the highest amino acid extraction conditions:2.23 times of extraction;extraction time is 61.94min;the best ratio of liquid and material 17.76mL/g.Optimal conditions for the extraction time is 62min;the best ratio of liquid and material 17mL/g;extraction times 2.23.3 In the process of analysis of free amino acids in Hovenia dulcis fleshy fruit stem,we get unknown component.Due to the use of derivatives and the high specificity of derivatization detection method,so we suppose this compound may be a new non-protein amino acid,or amino acid metabolic intermediates,or peptide etc.Because the unknown compound is detected by specific detection,we assume its physicochemical property is identical with amino acid residues,through the extraction process which is used on the separation and purification of amino acids.4 Preliminary separation and purification of unknown component of free amino acids in Hovenia fleshy fruit stalk.After running the NKA-2 macro porous resin and alcohol precipitation,we get the high purity exact of the unknown component.5 The purpose of the experiment is to figure out the influence caused by the extract of Hovenia dulcis and GABA on intestinal micro flora with the means of culturing dung mixed anaerobic in vitro.The result of the FISH count shows that GABA and Hovenia dulcis extract has a promoting effect on the growth of intestine micro flora,which significantly increased the content of beneficial bacterium Lactobacillus and Bifida bacterium,on the other hand,inhibition effect on harmful bacteria bacteria Clostridium is obvious but the same time,inhibition effect on the Bactericides has not been found clearly,but after 24 hours incubation,the content of the Bacteroides in fermentation liquid is lower than the control sample.There is no inhibition and promotion effect on the C.difficile bacteria.Through the detection of organic acid,lactic acid content was significantly higher than the blank control,this is due to the promotion effect.Besides,the total acid content was higher than that of the control.In this regard,we draw the conclusion:GABA and Hovenia dulcis extract can be exploited by the intestine,and generate short chain fatty acids. |
