As a new liquid dairy-based infant formula food,pasteurized infant formula milk is safe,convenient and nutritious compared with current infant formula food.At present,the infant formula food market in China is mainly based on infant formula milk powder,and infant formula milk is in its infancy in China.Ultra high temperature sterilization can result in loss of milk's own and added immune material activity,as well as partial protein denaturation.Among several heating treatments,pasteurization has the least effect on the denaturation of whey protein and can preserve the activity of immune substances in milk to the greatest extent.This paper aims to develop a pasteurized infant formula milk.According to the national food safety standard--infant formula food standard(GB 10765-2010),the optimum formula of pasteurized infant formula milk is designed and optimized by using Excel software and Matlab 7.0.The amino acid pattern,fatty acid pattern and trace element content of the formula meet the standards of infant formula food and close to breast milk.In order to ensure its safety,it is required to select the milk produced by healthy cattle with the lowest total number of bacteria as raw material,and use other sterilization technology to produce high-quality infant formula milk through pasteurization.The nutrition and efficacy of the product were verified by animal experiments.In this study,4-week-old SPF SD rats were fed with basic diet,commercial room temperature infant formula milk and commercial infant formula milk powder as control.We used ELISA to determine the effect of pasteurized infant formula milk on blood cytokines(IL-2?IL-4?IFN-??TNF-?)and lysozyme secretion level in rats,and flow cytometry to determine the effect of infant formula milk on T cell subsets.Compared with the basic diet group and the market infant formula group,pasteurized infant formula milk can significantly increase the body weight of rats,promote their growth and development,and significantly increase the secretion of c ytokines IL-2,IFN-?,TNF-?and lysozyme in rat serum,and increase the proportion of CD3+,CD4+,CD8+T lymphocyte in rat whole blood,and the ratio of CD4+/CD8+T lymphocyte is in the normal range.These results suggest that pasteurized infant formula milk ca n promote the growth of SD rats,promote the proliferation and differentiation of lymphocytes,and enhance the humoral and cellular immune functions.Based on 16 Sr RNA high-throughput sequencing technology to detect Alpha diversity,Beta diversity and phylum and genus level species composition of microorganisms in rat colon feces.The richness and uniformity of gut microbiota were improved in three groups of infant formula food.At the level of phylum,pasteurized infant formula milk significantly increased the relative abundance of Firmicutes and Actinobacteria in rats and decreased the abundance of Proteobacteria;at the generic level,it significantly increased the relative abundance of probioticssuch as Bifidobacterium and Lactobacillus,and decreased the r elative abundance of pathogenic bacteria such as Escherichia coli,Shigella and Klebsiella.This result is consistent with the trend of breast-feeding infant experiment.The results showed that the product could significantly promote the proliferation of intestinal probiotics,inhibit intestinal pathogens,and effectively improve the microecology of gut microbiota in rats.This study laid a scientific foundation for the practical production and application of pasteurized infant formula milk.
