| Metastasis inhibition plays a great significant role for treating cancers and improving the therapeutic effect of cancers.Functional factors existing in the diet and healthy eating patterns can effectively delay the development of cancers and inhibit cancer metastasis.However,at present,most of studies on the functional factors and dietary patterns for suppressing cancer metastasis focus on the results of suppressing metastasis,and the extremely limited researches had been done on the real-time monitoring of cancer metastasis.Moreover,the role of functional factors combined with dietary patterns on cancer metastasis has not been reported.The content of circulating tumor cells(CTCs)in peripheral blood is related to the stage of cancer development,which is of great significance for real-time monitoring of cancer metastasis.The CTCs in vivo capture method established by our research group can capture the CTCs in the peripheral blood of a breast cancer mouse model,combined with in vitro immunofluorescence staining,to obtain the change in the number of CTCs during the entire process of cancer metastasis,thereby achieving the real-time monitoring on the process of cancer metastasis,which could make up for the shortcomings that traditional methods can only obtain transfer results.Therefore,based on the in vivo capture method of CTCs,this paper had realized the real-time monitoring on the entire process of cancer metastasis effected by functional factors alone in breast cancer mouse models.And restriction of 30% dietary intake as a typical dietary pattern was selected,combined with functional factors,real-time monitoring of the combined effect of the two on the cancer metastasis process was conducted.In the first chapter,it was mainly summarized the functional factors and dietary patterns that are required to investigate their inhibitory effects of cancer metastasis and cancer development.Furthermore,the current assessment methods of inhibition cancer development and the detection methods of CTCs was summarized.In the second chapter,it was evaluated the effects of 11 types of functional factors on cancer metastasis based on the in vivo capture method of CTCs established in the laboratory,and compared the inhibitory effects of different functional factors on cancer metastasis in different stages of cancer metastasis.The experimental results were shown that different functional factors have different inhibitory effects on cancer metastasis at different time periods of cancer metastasis.Allicin and β-carotene had exerted the good inhibitory effects on metastasis throughout the cancer metastasis period.Resveratrol had exerted a good metastasis-inhibiting effect mainly in the early stage of cancer metastasis;while hesperidin,ganoderma lucidum polysaccharide,and catechin had played significantly inhibitory effects in the later stage of cancer metastasis.In the third chapter,based on the in vivo capture method of CTCs,it mainly explored the effects on the combination of three functional factors of allicin,hesperidin,astragalus polysaccharide and dietary restriction on the cancer metastasis process.Moreover,on the four aspects as the trend of changes in tumor volume in carcinoma-in-situ,mass in carcinoma-in-situ,amount of lung surface metastases and variation trend in CTCs,the impact of combined effects on cancer development was comprehensively evaluated.The experimental results were shown that the combined effect of different functional factors and dietary restriction has a certain degree of synergistic effect on suppressing cancer metastasis.In addition,based on the in vivo capture method of CTCs,when allicin was combined with dietary restriction,the whole cancer metastasis process was divided into the three stages with boundaries of the 17 th and 19 th day of tumor cell inoculation in the whole process of cancer metastasis.Before the 17 th day,the inhibitory effect of the combined effect group of allicin and dietary restriction on cancer metastasis showed a synergistic effect.Between 17 th and 19 th day,the inhibitory effect of combined action on cancer metastasis was between the individual effects of two factors.After the 19 th day,the inhibitory effect on cancer metastasis was shown as an antagonistic effect in the combined effect group.When astragalus polysaccharide and dietary restriction were combined.Since the 10 th day after tumor cell inoculation,the inhibitory effect of the combined effect on cancer metastasis had always located between the individual effects of astragalus polysaccharide and dietary restriction.This has certain reference value for choosing a healthy and reasonable diet for cancer patients. |
