Terminalia chebula Retz.is a traditional Chinese medicine,which is widely used in China,especially in Tibet and Inner Mogolia.Nowdays,currently research about it focuses on the chemical composition and pharmacology of some usual kinds of processed products,such as fried Terminalia,flour processed Terminalia,or bran processed Terminalia.However,there is no report about the chemical changes and special processing technology about the Rubia cordifolia L.processed T.chebula or Euphorbia fischeriana Steud processed T.chebula.Therefore,it is worth for further research.In this study,based on the "The basic processing technology in Tibet",the optimization of processing technology of two kinds of processed products were established,while the contents of moisture,ash and extract weight of that two kinds of special processed T.chebula were tested according to the relevant methods in "Chinese pharmacopoeia".And the contents of gallic acid,sennoside A,chebulinic acid were analyzed by using high performance liquid chromatography(HPLC),the contents of microelement were tested by flame atomic absorption spectrometry(FAAS).Furthermore,the TLC and HPLC fingerprint,as well as the optimization of processing technology of two kinds of processed products were established.And the chemical compositions of Euphorbia fischeriana Steud processed Terminalia chebula Retz.was studied by modern separation and detection methods.The main results were as follows:1.The optimization of processing technology for the two kinds processed productsWith the contents of gallic acid and ethanol extract as indexes,the processing technology of the two kinds of processed products were optimized by single factor and orthogonal experiments.The optimal results were as follows.?The Rubia cordifolia L.was added three times wheat wine after burning to white gray,which was stirred and boiled like buttermilk.Then the Terminalia chebula Retz.was added and soaked in it for 24h at room temperature.After the core was removed,the Rubia cordifolia L.processed Terminalia chebula Retz.was washed by distilled water for three times and dried at 70 ?for 30h.?The juice of the Euphorbia fscheriana Steud was obtained after boiling it in 15 times water.Then the same weight of Terminalia chebula Retz.was soaked in that juice for 48h at room temperature.After removing the core,the processed product was stewed in that juice again,and let it simmer until the juice dried up.Then the Euphorbia fscheriana Steud processed Terminalia chebula Retz.was died at 60 ? for 35h.2.Determination of the index for quality control of the different processed products of T.chebulaThe contents of moisture,ash and extract weight of the Rubia cordifolia L.processed T.chebula and the Euphorbia fischeriana Steud processed T.chebula were tested according to the relevant methods in "Chinese pharmacopoeia".The results showed that all the indexes were changed after the processing preparation.The contents of moisture varied from 6.6%to 9.19%and 7.96%,respectively,and the contents of total ash increased by 29.6%and 32.15%,respectively.The extract weight of that two kinds of special processed T.chebula increased to about 50%-60%.The contents of microelement were tested by FAAS,referring the contents of six kinds of microelements,including Fe?Mn?Co?Zn?Cr and Cu.And taking T.chebula as the control product,the contents of six kinds of microelements of the Rubia cordifolia L.processed T.chebula and the Euphorbia fischeriana Steud processed T.chebula were increased after processing.The contents of gallic acid,sennoside A and chebulinic acid were analyzed by using HPLC.Taking T.chebula as the control product,the contents of three kinds of chemical compositions of the Rubia cordifolia L.processed T.chebula and the Euphorbia fischeriana Steud processed T.chebula were changed after processing.The contents of gallic acid have increased by 25.33%and 64.32%,repectively.However,the contents of sennoside A decreased by 11.25%and 65.04%,respectively,and chebulinic acid reduced by 50.18%and 17.32%,respectively.On the other hand,the TLC fingerprint of triterpenes and polyphenol acids were established.And in the HPLC fingerprint,17 common peaks were found in Euphorbia fischeriana Steud processed T.chebula,while 21 common peaks were in Rubia cordifolia L.processed T,chebula.These results may offer evidence for the research of processing mechanism and provide theory basis for the quality control.3.The isolation and structure identification of compounds from Euphorbia fischeriana Steud processed T.chebulaIn this study,ten compounds have been isolated from Euphorbia fscheriana Steud processed T.chebula,including 2,4-dihydroxy-6-methoxy-3-formyl-acetophenone,2,6-dimethoxyphenol,Yuexiandajisu D,?-Sitosterol,24-methylenecycloartanol,2?,3?,19?-trihydroxy-12-oleanen-28-oic acid,arjunolic acid,arjungenin,2,4-dihydroxy-6-methoxy-3-methylacetophenone,20-twenty nine carbons olefine acid,and their structures were identified by MS,'H-NMR and 13C-NMR spectra.Among which,2,4-dihydroxy-6-methoxy-3-formyl-acetophenone,Yuexiandajisu D and 2,4-dihydroxy-6-methoxy-3-methylacetophenone were came from the Euphorbia fischeriana Steud.And 2,6-dimethoxyphenol and 20-twenty nine carbons olefine acid are isolated from this plant for the first time,which may be produced in the process of processing.
