Objective:These studies aimed to determine the contents of curcuminoids in Curcuma aromatica Salisb(CAS)extracted with ethanol,and to investigate the protective effects on myocardial ischemia and its acute and genetic toxicity.Methods:1?Curcuma aromatica Salisb was extracted with 30%,50%,70%and 90%ethanol.Rats in CAS groups were given extracts for 9 days.Acute myocardium ischemia models were established by pituitrin via intravenous and isoproterenol via subcutaneous injections.The changes of ST-segment and T wave of ECG and heart rate in different time periods were record after Pit and Iso injection.The the levels of myocardial creatinine kinase-MB(CK-MB)and lactate dehydrogenase(LDH),superoxide dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO)and total antioxidant capacity(T-AOC)in serum were measured.2?A total of 70 rats were randomly assigned to 7 groups(n=10/group):sham operation group,MI/RI model group,Danshen.Co group,curcumin group and CAS groups.Rats of CAS and danshen.Co group were perfused respectively with CAS and danshen.Co for 7 days beofore to ischemia/reperfusion.Other two groups were given NS instead of CAS equally before the surgery.The MI/RI model was established by ligaturing the left anterior descending branch of coronary artery(LAD)for 30 min and releasing the ligation later to reperfuse for 180 min in vivo.The activity of T-AOC,SOD,NO,CK-MB,LDH and MDA in serum were detected after reperfusing for 180 min.3?The contents of curcuminoids in Curcuma aromatica Salisb extracted with 30%,50%,70%and 90%ethanol were determined by HPLC.4?After the CAS was intragastrically administered one time at the maximal volume to mice in range of one day,the numbers of animals poisoned and died in each group were noted daily for 14 consecutive days.The maximum tolerated dose(MTD)was determined.The Ames tests using the Salmonella typhimurium strain TA97,TA98,TA100 and TA102 at five concentrations of 5,2.5,1.25,0.625 and 0.3125 mg/utensil of the CAS were carried out.The micronucleus tests(MNT)were implemented at three intragastrical doses of 25.6,12.8,6.4 g · kg-1 of the CAS.The tests of sperm aberration in mice were assayed at three intragastrical doses of 12.8,6.4,3.2 g · kg-1 of the CAS.Results:1?In the model group,compared with normal control group,acute myocardium ischemia model was successfully established by Pit and Iso.The rat heart rate was dropped rapidly,ST-segment and T wave of ECG were elevated and increased significantly(P<0.05).Levels of CK-MB,MDA and LDH in acute myocardium ischemia group were higher than those in normal control group(P<0.05);the activities of antioxidant enzymes SOD,T-AOC and NO were lower than those in normal control group(P<0.05).In CAS group,the rat heart rate,ST-segment and T wave of ECG were returned to normal levels in 15min,levels of CK-MB,MDA and LDH were lower than those in model group(P<0.05),activity of SOD,T-AOC and NO were higher than those in model group(P<0.05).According to the results of heart rate,ST-segment,T wave of ECG and the antioxidant index,the effects of CAS extracted with 70%ethanol on actue myocardial ischemia in rats were the most obvious.2?Compared with sham operation group,CK-MB in MI/RI group increased significantly(P<0.05),and the levels of MDA and LDH in serum were also higher than those in sham operation group(P<0.05),the activity of SOD,T-AOC and NO were decreased(P<0.05).3?The contents of curcuminoids were higher in Curcuma aromatica Salisb extracted with 70%and 90%ethanol than in Curcuma aromatica Salisb extracted with 30%and 50%ethanol significantly(P<0.05).4?The mean temperature difference(MTD)of CAS extracted with 70%ethanol was 25.6g ·kg-1,equal to the dried crude drug 197 g ·kg-1 and 300 times of clinical dose.The results of genetic toxicity test were negative.Conclusion:1?Curcuma aromatica salisb extracted with different ethanol' consistency show a good protection on IHD.According to comprehensive analysis,the effects of Curcuma aromatica salisb extracted with 70%ethanol on acute myocardium ischemia in rats are more obvious than others.All dose of Curcuma aromatica salisb have a good protection on MI/RI,but the effects in high dose group are more obvious.The protective mechanism of acute myocardium ischemia and MI/RI are not yet understood.We consider that Curcuma aromatica salisb protecte the heart against acute myocardium ischemia and MI/RI via reducing lipid peroxidation damage.2.The contents of curcuminoids in Curcuma aromatica Salisb were related to the extraction consistency of ethanol.3?According to the acute toxicity rating criteria,the extracts of CAS belong to non-toxicity and have no genotoxicity in the experimental condition.
