| Curcuma aromatica is the dry rhizome of Curcuma aromatica Salisb.,which is mainly grown in Guangxi Province,so it is also known as Guangxi Turmeric.Now it is contained in the “Zhuang Medicine Quality Standards” Volume II.Curcuma aromatica is warm in nature,pungent and bitter,and it is effect to promoting Qi flow and blood circulation,inducing menstruation to relieve menalgia.It is used for the treatment of sternocostal stinging,stomach pain,jaundice and epilepsy.As is one of the precious tranditional Zhuang medicine in our country,it has a long history and has accumulated definite clinical experience.Quality control is the key point to determine that whether the medicine can be used for clinical purposes.Therefore,our experiments study on the quality control of Curcuma aromatica,including herbal medicine identification,determination of moisture,total ash and ethanol-soluble extractives,content determination of four main known active ingredient,establishment of HPLC fingerprints of Curcuma aromatica and similarity evaluation,influencing factor test.The study is in order to provide science basis for the development and utilization of Curcuma aromatica in the future.1.According to the regulation of Curcuma aromatica in “Zhuang Medicine Quality Standards” Volume II,this experiment adopts TLC for thin layer identification of curcumin of 10 batches of Curcuma aromatica.The moisture,total ash and ethanol-soluble extractives of 10 batches were determined,and the testing indexes conformed to the regulation.Use HPLC for content determination of curcumin in Curcuma aromatica.The results show that the content of curcumin within the rules.2.TLC was used for thin layer identification of 10 batches of Curcuma aromatica.It detected the four medicinal materials containing ocathydro-1,4-dihydroxy-1,4-dimethyl-7-(propan-2-ylidene)azulen-5(1H)-one,curdione,aerugidiol.Selecting aerugi-diol,curcumin,procurcumenol,ocathydro-1,4-dihydroxy-1,4-dimethyl-7-(propan-2-ylidene)azulen-5(1H)–one,curdione as reference substance to establish the HPLC method for the content determination of five components in Curcuma aromatica.The methodological evaluation confirmed that the method is considered to be fast,simple,stable and reliable,good precision and reproducibility and it can provide a reference for quality evaluation of Curcuma aromatica and its preparations.3.To establish the HPLC fingerprints method of Curcuma aromatica,10 batches of Curcuma aromatica from different areas were detected,the similarity comparison and system cluster were applied to analysis HPLC fingerprints.The similarity comparison indicated that the similarities were all over 0.90.The 10 batches of Curcuma aromatica can be divided into 3 categories by the hierarchical cluster analysis.The experimental results indicated that the quality of 10 batches is stable,but the component content is different due to the collection,storage and other factors.It remains to be further research.4.Use the content variation of curdione of Curcuma aromatica under high temperature,light and moisture conditions after 5d and 10 d as an index for Influencing factor test.It can provide a foundation for development of Curcuma aromatica in the future and make it better to play the clinical effect. |
