Objective:To study the extraction technology of diterpenoid derived from Rabdosia excisa,and its protective effect on CCl4-induced acute liver injury in rats,and to explore its mechanism.Methods:In the experiment of extracting total diterpene from Kadsura alba,ethanol was used to extract ethanol and the content of propionin in Kochia scoparia was used as an index.The concentration of ethanol,the number of extraction and the extraction time were determined by orthogonal test The optimum selection of the total diterpene from Kadsura kamoji was studied.Rats in model group were given intragastric administration of CCl4 to induce acute liver injury in rats.The total diterpene extract of three kinds of low,CCl4 given intragastrically;normal control group given the same volume of blank solvent.To study the protective effects of total diterpenes from the leaves of Camellia japonicus on acute liver injury in rats by measuring the changes of serum ALT/AST/TBIL levels and liver histomorphology,and by measuring the indexes of oxidative stress,inflammatory cytokines and Mitochondrial membrane potential et al.Results:The preferred extraction process of the total diterpene from the leaves of Chayama japonica is:ultrasonic extraction with 75%ethanol for three times for 30 minutes.After intragastric administration of CCl4 for 24 h,obvious hepatic injury was observed in the rats.The serum biochemical parameters?ALT,AST and TBIL?and the changes of liver morphology?inflammatory cell infiltration,hepatocyte necrosis and hepatocyte swelling?.The total diterpene?100,200,500 mg/kg?extract of Kadsura longanae could significantly reduce the serum ALT,AST and TBIL levels induced by CCl4,and the total diterpene?500mg/kg?high-dose group can significantly reduce CCl4-induced liver pathological changes.In addition,the total diterpene extract from Kochia scoparia inhibits hepatic oxidative stress induced by CCl4,reversing the decrease of antioxidant enzymes induced by CCl4,partially blocking the production of free radicals,and also inhibiting GSH depletion and abnormalities of inflammatory cytokines Increased,effectively maintain the mitochondrial membrane potential and enhance mitochondrial MRC?,MRC?activity.Conclusion:Optimized Rabdosia excisa total diterpene extraction process is low cost and high efficiency.The total diterpene after caudate kaffir will significantly reduce the serum levels of ALT,AST and TBIL caused by CCl4,and at the same time it can improve the pathological changes of the liver and thus play a role in protecting the liver.Caudata fragrant Chaka may inhibit the oxidative stress,inhibit inflammatory cytokines,stabilize mitochondrial membrane potential to play anti-liver injury.
