Objective: The rat model of Ig A nephropathy was established by gavaging bovine serum albumin into rats,injecting castor oil and carbon tetrachloride subcutaneously,and injecting lipopolysaccharide into caudal vein.Huoxue Tongluo,Qingre Lishi traditional Chinese medicine was used to treat IgA nephropathy rat model.Observe the changes of 24 hours urinary protein,blood biochemical(such as total cholesterol,triglycerides,blood urea nitrogen and creatinine),renal pathological structure,renal tissue of rats with TGF-β1 mRNA expression and serum IgA1 concentration in order to investigate the mechanism of action and treatment effect of Huoxue Tongluo,Qingre Lishi traditional Chinese medicine.Methods: 40 SD rats weighing 180-200 g were reared in the experimental environment for one week.Then the rats were randomly divided into normal group,model group,benazepril group and traditional Chinese medicine group,10 rats in each group.Rats in model group,benazepril group and traditional Chinese medicine group were prepared by IgA nephropathy model.From the first to 12 th weeks,rats in model group,benazepril group and traditional Chinese medicine group were treated with BSA water solution 600mg/kg,and normal group rats were treated with distilled water 4ml/kg,once every other day;And rats in model group,benazepril group and traditional Chinese medicine group were subcutaneously injected with CCl4 0.1ml and castor oil 0.3ml,and the normal group was injected with normal saline 0.6ml once a week from the first to 12 th weeks.The rats in the model group,the benazepril group and the Chinese medicine group were injected with LPS 0.05 mg once a week after the tail vein in the 13 th and 14 th weeks,and normal group was injected with normal saline 0.2ml.From the 15 th to 18 th weeks,rats in the traditional Chinese medicine group were given Huoxue Tongluo,Qingre Lishi Recipe,benazepril group was given benazepril and the normal group and the model group were given the same amount of gastric.During the experiment,24 hours urine protein was detected.At the end of the experiment,rats were fasted for more than 12 hours,and injected with hydrated aldehydes at the abdominal cavity.The blood was collected at the femoral artery and the renal tissue was taken.The pathological changes of renal tissue were observed and the content of serum IgA1 and the expression of TGF-β1 mRNA were detectedResults:1.The rats in normal group were normal,which fur color were bright,reaction were sensitive,feeding and drinking water were normal.The model group,benazepril group and traditional Chinese medicine group rats were as general as normal rats before modeling.After making model,3 group rats gradually showed different degrees of listlessness,skin color or even worse hair loss,weight loss,slow reaction etc.The rats of benazepril group and traditional Chinese medicine group were treated with hydrochloric acid benazepril and Chinese medicine after intervention,the general situation of two groups rats was gradually improved.2.24-UTP: normal group,model group,western medicine group and Chinese traditional Chinese medicine group were all normal,with no statistically significant difference(P>0.05).After modeling,24-UTP in the model group,western medicine group and Chinese medicine group were significantly higher than the normal group,and the difference was statistically significant(P<0.05).To drug intervention treatment,western medicine,Chinese traditional medicine group 24-UTP were significantly lower in the model group,the difference was statistically significant(P<0.05),western medicine,Chinese medicine group is 24-there was no statistical significance(P >0.05)difference UTP.3.Urea nitrogen,creatinine: normal group,model group,western medicine group and Chinese medicine group,BUN and Scr were not significantly abnormal,and the difference was not statistically significant(P BBB 0 0.05).Total cholesterol,triglyceride,western medicine and Chinese medicine group rats total cholesterol,triglycerides,there were no obvious changes were compared with normal group,there was no statistically significant difference(P>0.05),the model group rats triglycerides no significant change in the normal group,there was no statistically significant difference(P>0.05),total cholesterol to higher than normal group,the difference was statistically significant(P<0.05);The normal group,western medicine and Chinese medicine group rats triglycerides are no significant change in the model group,there was no statistically significant difference(P>0.05),total cholesterol are down,and compared with model group the difference was statistically significant(P < 0.05).4.Renal histopathological observation.Light microscopy: the size and morphology of glomeruli in normal rats were normal,and no obvious thickening was observed in the mesangium.The glomerular morphology of rats in the model group was normal and the membrane was thickened.In the western medicine group and the traditional Chinese medicine group,the glomerular morphology was normal,and the membrane was thicker than the normal group,and the pathological changes of the rats in the model group were less.Immunofluorescence: normal group rat glomerular mesangial area did not see the immune complex deposition,glomerular mesangial area visible model group rats green fluorescent complex deposition,western medicine,Chinese traditional medicine group rat glomerular mesangial area all have different degrees of fluorescent complex deposition,rats in the model group.5.RT-PCR was used to observe the expression of TGF-β1 1 mRNA in renal tissue.In the model group,western medicine group and Chinese medicine group,the expression of TGF-β1 mRNA was significantly higher than that in the normal group,and the difference was statistically significant(P<0.05).The expression of TGF-β1 mRNA in Chinese medicine group and western medicine group was significantly lower than that in the model group,and the difference was statistically significant(P<0.05).Compared with the traditional Chinese medicine group and the western medicine group,there was no significant difference in the expression of TGF-β1 m RNA,and the difference was not statistically significant(P>0.05).6.he content of serum IgA1 was observed by ELISA.The contents of serum IgA1 in the model group,western medicine group and Chinese medicine group were significantly higher than that in the normal group,and the difference was statistically significant(P<0.05).The serum IgA1 contents in the Chinese medicine group and the western medicine group were all significantly lower than those in the model group,and the difference was statistically significant(P<0.05).There was no significant difference in serum IgA1 content between Chinese medicine group and western medicine group,and the difference was not statistically significant P>0.05).Conclusion:1.Huoxue Tongluo Qingre Lishi traditional Chinese medicine can improve the general situation of IgA nephropathy rats,decrease the 24h-UTP iprotein quantitative,down regulate the content of IgA1 and the expression of TGF-β1 mRNA,relieve renal pathological damage and protect renal function,delay the course of IgA nephropathy,and provides experimental basis for Huoxue Tongluo Qingre Lishi traditional Chinese medicine in the treatment of IgA nephropathy2.The reason how Huoxue Tongluo Qingre Lishi traditional Chinese medicine down regulating the expression of TGF-β1 mRNA may be associated with down-regulation of content of IgA1 in serum.But the sample size in this study is less,the future still need a large sample of experimental research...
