| Objective The expression level of carcinoembryonic antigen cell adhesion molecule 6 oncoprotein and matrix metalloproteinase 9 oncoprotein were both tested in breast cancer tissues and the nomal adjacent breast tissues by using Immunohistochemistry improved streptavidin-perosidase(S-P)technique,besides,the clinical significance was discussed,and theoretical basis for monitoring development of prognosis and targeted therapy was provded.Then we can make a better understanding about breast cancer.Methods Sixty samples of breast cancer and corresponding adjacent normal breast tissue were randomly collected in the 1st affiliated hospital of Bengbu medical college from January first,2013 to January first,2015.Immunohistochemistry improved streptavidin-perosidase(S-P)technique was used to detect the expressions of CEACAM-6 and MMP-9 oncoprotein in above-mentioned 60 cases of breast cancer tissues and corresponding adjacent normal breast tissues.When the experiment came to an end,the date of the immunohistochemistry and corresponding clinical were retrospective analyzed by using SPSS 20.0 statistical package.We used chi-square test to inspect the correlation among the expression of CEACAM-6、MMP-9 in the age,gender,TNM stages,grades of differentiation,metastasis of lymph nodes.And Spearman rank correlation analysis was used to analyze the relativity of CEACAM-6 and MMP-9 oncoprotein.Moreover,the immunohistochemistry results of all cases in ER,PR,HER-2,Ki-67 receptors were collected.We analyzed the relationship between CEACAM-6 and HER-2 receptor,as well as MMP-9 oncoprotein,and the relationship among different molecular subtypes of breast cancer were did too.Results 1 The positive expression rate of CEACAM-6 oncoprotein in breast cancer tissue was 61.7%,howerve,the positive rate of CEACAM-6 oncoprotein in corresponding adjacent normal breast tissue was 38.3%.There was significantly difference between the two date and it was in accord with medical statistics(P<0.05).2 The positive expression rate of MMP-9 oncoprotein in group of corresponding adjacent normal tissues of breast cancer was 18%,on the contrary,the positive rate in breast cancer tissues was 78%.And There was significantly difference between the two date and it was in accord with medical statistics too(P<0.05).3 By using Spearman rank correlation analysis,we have authenticated that the expression of CEACAM-6 was positively correlated to MMP-9 oncoprotein in above 60 cases.They may contribute to the occurrence,invasion and metastasis of the breast cancer.4 By using chi-square test,we approved that neither tumor size nor age was associated with the expression of CEACAM-6 and MMP-9 in mammary cancer.And the lymph node metastasis,TNM stages and grades of differentiation were all associated with the two oncoprotein in contrast.5 The expression of CEACAM-6 and MMP-9 in different molecular subtypes of breast cancer was not the same,and the two oncoprotein were especially positive in Her-2 overexpression subtypes.Besides,the two oncoprotrin were positively correlated to HER-2 receptor.It was in accord with medical statistics(P<0.05).Conclusion 1 The expression of CEACAM-6 in corresponding adjacent normal breast tissue was more negative than that in mammary cancer.2 Contrasting the breast cancer tissue and corresponding adjacent normal breast,we have investigated that the rate of MMP-9 was more higher expressed in the former.3 Neither tumor size nor age was related to the expression of CEACAM-6 and MMP-9 oncoprotrin in mammary cancer,on the contrast,all of lymph node metastasis,TNM stages and grades of differentiation were associated with them.4 CEACAN-6 and MMP-9 may contribute to the occurrence and development of the breast cancer,because they were positively correlated expressed in above 60 samples and their relationship had medical statistical significance.5 Different molecular subtypes of breast cancer had different expression level of CEACAN-6 and MMP-9,the rate of Her-2 overexpression subtypes was especially higher than the other three,and their difference had medical statistical significance in above 60 cases.Moreover,CEACAN-6 and MMP-9 were positively correlated to HER-2 receptor.To summarize,moderate and strong expression of CEACAM-6 and MMP-9 oncoprotrin in mammary cancer may cause invasion and metastasis,and joint detection of CEACAM-6 and MMP-9 may have an important role to forecast the occurrence,development and treatment effect. |
