Experimental Study On Effect Of Bushenhuoxue Fang On Anticardiolipin Antibody Positive Serum Mouse TNF-a,COX-2 Effect

Objective:To further explore the mechanism of Bushenhuoxue treatment of autoimmune abortion,using recombinant human ACA positive animal model was established by intraperitoneal injection of β 2GP I,observing the influence of Bushenhuoxue ACA cardiolipin antibodies inserum of mice,anti-β2GP I TNF-a,COX-2.Dicovering the relationship between TNF-α,COX-2 cytokines and anti-cardiolipin antibodies recurrent abortion preliminary,providing a basis for the study of anti-cardiolipin antibodies recurrent miscarriage signal transduction pathways,laying the foundation for further research.Methods:BALB/c mice using recombinant human ACA positive animal model was established by intraperitoneal injection of β 2GP I,48 BALB/c mice were randomly divided into Bushenhuoxue high,medium and low dose group,medicine(aspirin)group,model group,the control group-a total of 6 groups,8 each group,marked after weighed and recorded as corresponding group A,group B,group C,D group,E group,F group.Grouping administration:By discovering vaginal suppository or vaginal smear under the microscope,finding a large quantity of sperm recorded as the first day of pregnancy,according to the formula "mice per kg bw=body weight in humans generally dose X(mouse dose conversion factors/human dose conversion factor)"calculated mice fed doses,gavaged 14 days as the calculated dose:on 15th day of pregnancy,each group of mice to be blood,put to death,stripped the uterus and marked,then calculated the number of stillbirths.ELISA assay Bushenhuoxue and the remaining five groups of mice serum ACA,anti-β 2-GP I antibody,TNF-a and COX-2 levels.Results:1.This study successfully established an animal model of anti-cardiolipin antibodies in mice.2.In these three groups-Bushenhuoxue high,medium and low dose group,the medium is most significant.Bushenhuoxue medium dose group and western medicine aspirin group comparison P>0.05,not statistically significant,adjust model mice serum ACA,anti-β 2-GP I antibody levels close to the control group3.Bushenhuoxue can reduce abnormally elevated tumor necrosis factor in serum,elevate reduced cyclooxygenase 2,make an overall improvement in peripheral blood and the local environment within the uterus,regulate the body’s immune imbalance state.
