Jujube leaf is a part of jujube,which is often discarded as the inedible part,the purpose of this paper is to make full use of resources to improve the utilization rate of jujube trees,the dynamic changes of the bioactive substances in different jujube leaves were measured;the content of total flavonoids and total phenolics in different leaves were determined,and the antioxidant activity was determined;the extraction,separation,purification and structural analysis of jujube leaves were carried out by column chromatography and liquid chromatography;establishment of HPLC fingerprints of jujube leaves;the anti-aging test of total flavonoids from jujube leaves was studied by a series of anti-aging and anti-aging experiments in vitro and in vivo.The results are as follow:1.The dynamic changes of total flavonoids and total phenolics in leaves of Zizyphus jujube cv.Junzao,Zizyphus jujube cv.Huizao,sour jujube showed a series of fluctuating trends from high to low,the total content of total flavonoids was the highest at flowering stage,and the total phenolics content of Zizyphus jujube cv.Junzao,Zizyphus jujube cv.Huizao jujube leaves reached the highest level in fruit ripening period,the total triterpenes and total saponins showed an increasing trend and also reached the highest level in fruit ripening period;the antioxidant ability of jujube leaves was weak in fruit ripening stage;the content of rutin in four flavonoids was the highest,it was 13.37±0.61 mg/g,followed by hesperidin,Scutellarin,Naringin,Kaempferol,Quercetin.2.The content of total flavonoids and total phenolics in 45 varieties of jujube leaves were different,the highest and lowest levels of flavonoids were Zizyphus jujube cv.Binglang and Zizyphus jujube cv.Sanbianse,the contents were 41.41±2.81 mg/g,9.26±0.55 mg/g;the highest and lowest levels of total phenolics were Zizyphus jujube cv.Zhongzao 3 and Zizyphus jujube cv.Zan 2,the contents were 214.19±3.83 mg/g,68.14±0.63 mg/g;the antioxidant ability was strong is the Zizyphus jujube cv.Mohu,the weakest is the Zizyphus jujube cv.Sanbianse;by cluster analysis,the 45 jujube varieties are divided into five categories,the first group gathered 12 kinds of jujube leaves,the second group gathered six kinds of jujube leaves,the third category gathered 17 kinds of jujube leaves,the fourth group gathered eight kinds of jujube,Five species were gathered two kinds of jujube.3.Seven compounds were isolated from the ethanol extract of Zizyphus jujube cv.Huizao jujube leaves,which were quercetin?1?,rutin?2?,kaempferol-3-O-rutin?3?,kaempferol?4?,isoquercitrin?5?,Astragalin?6?,7-hydroxy-6-methoxycoumarin?7?;?1?6is a flavonoid compound,and?7?is a coumarin compound.4.The fingerprints of 29 kinds of jujube leaves were established by high performance liquid chromatography,and 14 common peaks were calibrated,the relative time and the relative peak eare of the RSD values were than 5%,29 kinds of jujube leaves could be divided into two grades by the fingerprints of traditional Chinese medicine.5.70% ethanol extract of jujube leaves has strong antioxidant activity,artificial gastric juice on jujube leaves after treatment,the antioxidant activity has increased significantly,artificial intestinal juice on jujube extract treatment,the antioxidant activity has increased,and the results showed that the flavonoids of jujube leaves could increase the body weight of aging mice,the liver coefficient and heart coefficient of aging mice,the positive control and the different dose of D-galactose of the test substance in heart,liver,serum can play a different antioxidant and anti-aging effect,enhance the SOD activity,reduce the accumulation of harmful substances such as peroxide,improve the aging caused by the mouse heart,liver,kidney pathology changes,improve the various aspects of mouse physiological function.
