Objective: To further improve biological activity of medical degradable magnesium,human lactoferrin(HLF)was loaded into pure magnesium ultrasonic microarc oxidation(UMAO)-silane film surface,the effects of different concentrations of HLF on cell membrane compatibility were studied and a theoretical basis for its orthopedic implant was provided.Methods: The specimens were divided into four groups.the pure magnesium UMAO-silane was group A,pure magnesium UMAO-silane-0.05 mg / ml HLF,pure magnesium UMAO-silane-0.1mg / ml HLF,pure magnesium UMAO-silane-0.2mg / ml HLF were respectively group B,group C and group D.The surface morphology of the film was observed by scanning electron microscope(SEM),film elements was analysised by energy spectrum analyzer,the performance of film was studied by wetting angle meter.Cell adhesion and cell proliferation were studied by living cell counting kit(CCK-8),the number and morphology of cells were observed by laser confocal microscope.Results: SEM in group B,group C,group D closed microporous membrane layer effect was remarkable,The pore diameter and number of micropore were decreased compared with group A.Energy spectrum analysis of membrane layer of MgO as the main component of group A,The content of N element in the film layer of group B,C and D was significantly higher than that of group A,and the Si element content was significantly decreased..The value of wetting angle was group A > group D > group C> group B.in the cell adhesion and proliferative experiments,the optical density(OD)value determined by the automatic enzyme marker,the OD value of the pure magnesium UMAO-silane-0.1mg/ml HLF film layer was the largest.At the same time,the OD value of each group increased with time.The statistical analysis of each group was P<0.05.Confocal microscopy images showed that the pure magnesium UMAO-silane-0.1mg / ml HLF layer had the largest number of cells and the best cell stretch.Conclusions: Pure magnesia UMAO-silane-HLF composite membrane layer has some degree of pore filling and hydrophilic function,The cell adhesion and proliferative ability of pure magnesium UMAO-silane-0.1mg/ml HLF were the best.
