Objective:To study the effects of four kinds of drinking water on immune function in mice,including pure water,tap water,filtered water and mineralized water.Methods:Firstly,water quality of four kinds of drinking water,including pure water,filtered water,tap water and mineralized water were analyzed.The index contained general parameters,cationic and anionic parameters.Then a series of animal experiments were conducted for detecting immune function of ICR mice by drinking four kinds of water freely.Totally four batches of ICR mice,every one batch containing 80 mice(half male and half female)were randomly divided into four groups by weight,drinking pure water,tap water,filtered water and mineralized water respectively for 90 days.Nonspecific immune function(carbon clearance test,peritoneal macrophage phagocytosis test,NK cell activity test by LDH assay),the immune organ index(thymus coefficient,spleen coefficient),cellular immune function(spleen lymphocyte transformation test,delayed type hypersensitivity),humoral immunity(half hemolysis test,detecting immunoglobulin G and M by ELISA),and testing T-lymphocyte subets by flow cytometry,cytokine detection for IL-2,IL-4,IL-6,,IL-10,IFN-? and TNF-? with ELISA,white blood count,neutrophil count,monocyte count,were studied for evaluating the immune effects of four kinds of water on mice.Results:1.Analysis of four kinds of drinking water quality: pure water has almost no mineral elements.The mineralized water is alkaline,and the contents of copper and magnesium are higher than that of pure water.Both filtered water and tap water,with higher TDS and EC,contain more cations and anions than pure water and mineralized water.The filter water is alkalescent,with negative ORP and contains a certain amount of dissolved hydroge.2.Compared with each group,carbon clearance phagocytosis index a was not different(P>0.05).Peritoneal macrophage phagocytosis rate of filter water group was the highest,comparing with mineralized water group(P<0.05),pure water group(P>0.05)and tap water group(P>0.05).Peritoneal macrophage phagocytosis index of filter water group was significant higher than pure water and mineralized water(P<0.05).NK cell activity in filter water group was obviously higher than pure water group and mineralized water group(P<0.05),even higher than tap water group(P>0.05).3.Compared with each group,there were not significant differences in rised weigh,thymus coefficient and spleen coefficient,ear swelling degree,splenocyte proliferation(P>0.05).4.Compared with each group,there were not significant differences in serum half hemolytic reaction,Ig G and Ig M(P>0.05).5.Compared with each group,there were not obvious differences in CD4+and CD8+percentage of T cell subsets,IL-2,IL-4,IL-6,IL-10,IFN-? and TNF-?,white blood count,neutrophil count,monocyte count(P>0.05).CD4+ /CD8+ rate of tap water group,filtered water group and mineralized water group were significant higher than pure water group(P<0.05).Conclusions:Drinking filtered water for a long time could promote the non-specific immune function of mice by enhancing the phagocytosis of peritoneal macrophages and increasing the activity of NK cells.Filtered water and mineralized water could increase the ratio of CD4+/ CD8+in spleen cells of mice.
