Objective : To investigate the correlation between the two MGMT gene loci35MGMT(66G>A)(rs16906252)and 34MGMT(2661500A>G)(rs531572)in peripheral blood of patients with glioma after operation and the chemotherapy of glioma.Method:A retrospective analysis was made on 65 patients with glioma who had been operated on for the first time in our hospital from January 2017 to July 2018.The expression of MGMT in peripheral blood was detected by digital fluorescence molecular hybridization and Pearson correlation grade was analyzed with 1p/19 q deletion and IDH1 mutation.Results:Among65 patients with glioma,35MGMT(66G>A)(rs16906252)was found in 0 cases of methylation(AA,AG),and all of 65 cases were GG type.Among 34 MGMT(2661500A >G)(rs531572)genotypes,13 were AA,36 were AG and 16 were GG.The expression of MGMT protein was not correlated with tumor grade,sex and age(P=0.075,P=0.193,P=0.335).Moreover,Pearson correlation test showed that the correlation coefficients of MGMT expression with 1p/19 q combined deletion and IDH1 mutation were ? =-0.708(P<0.001<0.05)and ?=-2.178(P=0.029<0.05)respectively.namely,In patients with 1p/19 deletion and IDH1 mutation,the expression of MGMT was low and TMZ was sensitive to chemotherapy.Conclusion: Detection of this gene locus in peripheral blood of glioma patients(MGMT rs16906252)is not necessarily suitable for Chinese,and this conclusion still needs to be confirmed by follow-up studies.There was no difference in the chemosensitivity of TMZ among gliomas of different grades,and there were gliomas with 1p/19 q deletion and IDH1 mutation deletion.TMZ had high chemosensitivity.This gene locus(MGMT rs531572)may plays a guiding role in the individualized application of TMZ after surgery for glioma.
