Study On The Quality Evaluation Of Buleuri Radix And Its Decoction Pieces

Objective:By studying the correlation between the appearance and internal indexes of Buleuri Radix and its Decoction pieces,the quality of Buleuri Radix and its decoction pieces was evaluated,and the multi-index,multi-component and comprehensive quality standards and grade standards were established,which laid the foundation for the formulation of industry standards.Methods:1.Collect samples of Buleuri Radix materials and decoction pieces from the real estate area,main producing area,mainstream market,and distribution center,and carry out the identification of the base source.Refer to the 2015 edition of the Chinese Pharmacopoeia and the Hong Kong Chinese herbal medicine standard and other related medicinal materials standards for the samples.Source,traits,identification,inspection,fingerprints,extracts,content determination and other projects were carried out to study,to establish new quality standards for Buleuri Radix materials and decoction pieces,and to evaluate the quality of Buleuri Radix medicinal materials and decoction pieces.2.Drawing on the existing medicinal materials and decoction pieces grading standards,combined with market circulation,clinical application and grading,according to experimental measurements of the diameter of the thick end,fine end diameter,middle circumference,length,color and other appearance indicators,moisture,total ash Intrinsic indicators such as acid-insoluble ash,leachate,and saikosaponin content were analyzed by correlation analysis,principal component analysis and cluster analysis to investigate the correlation between indicators and establish a grade evaluation standard for Buleuri Radix medicinal materials.3.By examining the appearance traits,volatile oil,extracts,saikosaponin a,d content of traditional Chinese medicine decoction pieces,the appearance traits and internal indexes of Buleuri Radix decoction pieces were quantified,and the composite mass constant was constructed to form a Chinese medicine with a clear numerical range.The product specification grade of the decoction pieces is perfected and the grade standard of Buleuri Radix decoction pieces is established.Results:1.Collect 48 batches of medicinal materials and 20 batches of decoction pieces.The appearance traits,moisture,ash inspection,leachables and active ingredients are all in line with the Chinese Pharmacopoeia 2015 edition standard.However,the study found that the Buleuri Radix cultivars were chaotic,and different Buleuri Radix cultivars required thin layer identification,fingerprinting and identification of base sources.2.Based on the existing standards,the method of thin layer identification was optimized and the method of microscopic identification,volatile oil and fingerprint of Buleuri Radix was established.Buleuri Radix medicinal grade standard:Through correlation analysis,the total amount of extract and the color of the medicinal material L~*,the color of the medicinal material a~*,the color of the medicinal material b~*,and the total amount of saikosaponin a and saikosaponin d are extremely significant.Correlation,the fine-end diameter of Buleuri Radix was significantly positively correlated with the b~*of Buleuri Radix.The total ash and acid-insoluble ash of Buleuri Radix,the color of the drug a~*,and the total amount of saikosaponin a and saikosaponin d were Significantly positive correlation.Principal component analysis was used to determine the content of saikosaponin a and saikosaponin d,and the extract was the main intrinsic index for evaluating the quality of Buleuri Radix.The color of acacia medicinal herb a~*,the color of bupleurum b~*,the diameter of the fine end as the main indicator.The clustering analysis shows that the grades are as follows:First class of Buleuri Radix:the diameter of thin end was more than 2.32 cm,chromaticity a~*of medicinal materials was more than 12.83,chromaticity b~*of medicinal materials was more than35.59,the content of saikosaponin a and saikosaponin d were more than 4.24%,the content of extract was more than 36.38%.Second class of Buleuri Radix:the diameter of thin end was 1.96~2.32 cm,chromaticity a~*of medicinal materials was 9.79~12.83,chromaticity b~*of medicinal materials was 28.77~35.59,the content of saikosaponin a and saikosaponin d were 1.05%~4.24%,the content of extract was 17.71%~36.38%.Third class of Buleuri Radix:the diameter of thin end was lower than 1.96 cm,chromaticity a~*of medicinal materials was lower than 9.79,chromaticity b~*of medicinal materials was lower than 28.77,the content of saikosaponin a and saikosaponin d were 0.30%~1.05%,the content of extract was 11.00%~17.28%.3.Classification standard of Buleuri Radix decoction pieces:the compound quality constant of first grade was≥7.871;the second grade was<7.871 but≥5.602;while the third grade was<5.602.There is no clear division requirement for this method,and it can be changed according to actual needs.Conclusions:1.Studies have shown that Buleuri Radix varieties are confusing,and different Buleuri Radix varieties need to be identified by thin layer,fingerprint and base source identification.2.The newly established Buleuri Radix quality standard supplements the limits of the unspecified items of the 2015 edition of the Chinese Pharmacopoeia,establishes new quality evaluation standards for Buleuri Radix medicinal materials and decoction pieces,and lays the foundation for the formulation of industry standards.3.For the first time,mathematical statistics and composite mass constant methods were used to establish standards for the classification of Buleuri Radix medicinal materials and decoction pieces,which laid the foundation for standardizing the quality control of medicinal materials and decoction pieces and realizing high quality and good prices of medicinal materials and decoction pieces.
