Objectives In this study,Wistar rats were fed with high-fat diet to construct the model of insulin resistance obesity rats,and electroacupuncture was used for intervention.The effects of electroacupuncture on Ap N,Leptin and Resistin in white adipose tissue were observed,and the related effects and mechanism of electroacupuncture on improving insulin resistance obesity were discussed.Methods Will only 8 weeks of age 60 male Wistar rats by giving ordinary adaptive feed feed a week later,the randomly divided into normal group and 12 feed and high fat feedstuff group and 48,among which gives ordinary ordinary feed feed,high fat feedstuff group was given high fat feed,two groups were fed 8 weeks,then respectively from ordinary feed group,high-fat feed group of randomly selected from 6,24 Wistar rats line high insulin and glucose clamps,used for assessment of insulin resistance,calculate the Lee of the two groups of rats' s index,used to assess obesity.After the successful modeling,24 insulin resistant obese rats in the high-fat diet group were randomly divided into model control group,electro-acupuncture intervention group,sham electro-acupuncture intervention group and resveratrol group,6 rats in each group.The remaining six rats in the normal feed group were named as the normal control group.Among them,the normal control group was only given ordinary feed,without any other experimental measures;The model control group was fed with high-fat diet without any other experimental measures.Electric acupuncture intervention group was given a high-fat feed,at the same time give the curative intervention,select "st 36" "chung wan","hong" pivot "day" four holes,connect Korean after acupuncture cupping apparatus,selection of frequency of 2 hz,the intensity is 1 ma continuous wave,10 minutes per treatment,treatment once a day,and treatment of 7 times a week,the total course of 8 weeks,around the pivot foot three mile,hong leong,day used interchangeably.After 8 weeks of intervention,the body mass of the above 5 groups of rats was measured and the Lee's index was calculated.Hyperinsulin-normal glucose clamp was used to determine the glucose infusion rate(GIR).Blood was collected from abdominal aorta and serum FFA,TC and TG levels were detected by ELISA.And then remove the white adipose tissue,using Western blot method(Western Blotting,WB)detection Ap N,Leptin,Resistin protein expression levels;Finally,the whole data is analyzed and processed by SPSS software.Results1.Effects on rat weight,Lee's index and GIR after high fat feeding for 8weeks The weight and Lee's index in high fat feeding group were significantly higher than normal group(P<0.01),and GIR in high fat feeding group were significantly lower than normal group(P<0.01)after feeding of eight weeks.2.Effects on rat weight,Lee's index and GIR after electricacupuncture intervention for 8 weeks The body weight and Lee's index of the model group were significantly higher than those of the normal control group(P<0.01).The body weight and Lee's index of the model group were significantly higher than those of the electroacupuncture intervention group and the resveratrol group(P<0.01).The body weight and Lee's index of the rats in the non-electroacupuncture intervention group showed a downward trend,but compared with the model control group,no statistical significance was found between the model control group and the non-electroacupuncture intervention group(P>0.05).The body weight of the electroacupuncture intervention group was significantly lower than that of the non-electroacupuncture intervention group(P<0.05).Although the body weight of the electroacupuncture intervention group was lower than that of the resveratrol group,there was no statistical significance between the two groups(P>0.05).GIR values of the rats in each group were significantly lower than those in the normal control group(P<0.05).The model control group was significantly lower than the electroacupuncture intervention group and the resveratrol group(P<0.05).The electroacupuncture intervention group was significantly higher than the non-electroacupuncture intervention group(P<0.05).There was no significant difference between the non-electroacupuncture intervention group and the model control group(P>0.05).Although the electroacupuncture intervention group was slightly higher than the resveratrol group,there was no statistical significance between the two groups(P>0.05).3.Effects of electricacupuncture on FBG,PBG,the level of FFA,TC,TG in insulin resistance in obese rats Comparison of serum TC,TG and FFA levels in rats of each group:the levels in the model control group were significantly higher than those in the normal control group(P<0.05).The electroacupuncture intervention group and resveratrol group were significantly lower than the model control group(P<0.05).Serum TC,TG and FFA levels in the electroacupuncture group were lower than those in the resveratrol group,but there was no significant difference between the two groups(P>0.05).There was no significant difference between the non-electroacupuncture intervention group and the model group(P>0.05).The electroacupuncture intervention group was lower than the non-electroacupuncture intervention group(P<0.05).4.Effects of electricacupuncture on the level of protein of Ap N?Leptin?Resistin in insulin resistance in obese rats.Compared with the normal control group,APN protein expression level in the model control group was higher,and there was no statistical significance between the two groups(P > 0.05).The expression level of APN protein in the model control group was significantly lower than that in the electroacupuncture intervention group,the non-electroacupuncture intervention group and the resveratrol group,with statistical significance(P<0.05).The expression level of APN protein in the electroacupuncture intervention group was higher than that in the resveratrol group,and the comparison between the two groups was statistically significant(P<0.05).The APN protein expression level in the electroacupuncture intervention group was higher than that in the non-electroacupuncture intervention group,and there was no statistical significance between the two groups(P>0.05).Compared with the normal control group,the Leptin protein expression level in the model control group was significantly increased(P<0.05).The Leptin protein expression level in the model control group was significantly higher than that in the electroacupuncture intervention group,the non-electroacupuncture intervention group and the resveratrol group,with statistical significance(P<0.05).The Leptin protein expression level in the electroacupuncture intervention group was lower than that in the resveratrol group,and the comparison between the two groups was statistically significant(P<0.05).Leptin protein expression level in the electroacupuncture intervention group was lower than that in the non-electroacupuncture intervention group,and the comparison between the two groups was statistically significant(P<0.05).Compared with the normal control group,the protein expression level of Resistin in the model control group increased,but there was no statistical significance(P > 0.05).The expression level of Resistin protein in the model control group was significantly higher than that in the electroacupuncture intervention group,the non-electroacupuncture intervention group and the resveratrol group,with statistical significance(P<0.05).The protein expression level of Resistin in the electroacupuncture intervention group was lower than that in the resveratrol group,and the comparison between the two groups was statistically significant(P<0.05).The protein expression level of Resistin in the electroacupuncture intervention group was lower than that in the non-electroacupuncture intervention group,and the comparison between the two groups was statistically significant(P<0.05).Conclusions1.The body weight,Lee's index,the level of TC,TG,FFA,FBG and PBG can be reduced by electricacupuncture,meanwhile,insulin resistance of the insulin resistance in obese rats can be improved.2.Electroacupuncture can promote the expression of APN and reduce the expression of Leptin and Resistin proteins in the white adipose tissue of insulin-resistant obese rats,so as to promote the differentiation of adipocytes,improve lipid metabolism,regulate energy metabolism and improve the status of insulin resistance obesity.
