[Objectives] To characterize Th17-cell immune responses to Helicobacter pylori(H.pylori)infection,and to explore the role and pathology mechanisms of Th17-cell in the H.pylori infection.[Methods] 1.Chemokine and cytokine related to Th17-cell in the serum from patient with H.pylori infection were determined by ELISA.2.C57BL/6 mice were infected with H.pylori orally.Serum,spleen and gastric tissues were collected at the indicated times post-infection.The frequency of Th17 cells of spleen and gastric tissue were determined by flow cytometry.The concentrations of CCL20,IL-17,IL-1?,TNF-?,and IL-6 were tested by ELISA.3.Bioinformatic analysis were applied to explore hub genes and potential biomarkers of H.pylori infection.[Results] 1.Both CCL20 and IL-17 were expressed at a higher level in serum from H.pyloriinfected patients.2.In H.pylori-infected mice model,Th17-cell in spleen and in gastric tissues were expanded on day 14 post-infection and peaked on day 21 post-infection.The concentration of CCL20 in gastric tissues peaked on day 7 post-infection.The level of IL-17 in serum peaked on day 14 post-infection,whereas in gastric tissue on day 21 postinfection.The concentration of IL-1?,TNF-?,and IL-6 were positive correlated with IL-17.3.CXCL8,CXCL1,CXCL10,CXCL6,CXCL5,and CCL20 were chosen as potential biomarkers for treatment of H.pylori-infection by bioinformatic analysis.[Conclusion] 1.H.pylori-specific Th17 immune responses were elicited in both human and mice model.2.Th17-cell related cytokine IL-17 may facilitate the expression of inflammation cytokines.The role of IL-17 on H.pylori infection may be associated with the expression of IL-1?,TNF-?,and IL-6.
