| Objective:By detecting the expression of survivin protein and survivin m RNA and the frequency of apoptotic cells in nasal mucosa CD4~+ T cells in patients with seasonal allergic rhinitis(SAR),investigate the effect of survivin on apoptosis of CD4~+ T cells in nasal mucosa of SAR patients and the potential regulatory mechanisms of the process.Methods:Collecting 30 cases of SAR patients(SAR group)from the Department of Otolaryngology,Second Hospital of Shanxi Medical University from January 2016 to January 2018 and 30 cases of simple nasal septum(control group)in the inferior turbinate mucosa,grinding and collecting mononuclear cells.CD4~+ T cells were isolated by density gradient centrifugation,the expression levels of survivin protein and survivin m RNA in CD4~+ T cells were detected by Western-blotting and RT-qPCR respectively.Flow cytometry was used to detect the frequency of apoptotic cells after treatment of CD4~+ T cells with AICD activator.To investigate the effect of survivin on the apoptotic cells of CD4~+ T cells in nasal mucosa of AR patients.Results:The level of survivin protein in CD4~+ T cells in nasal mucosa of SAR group was significantly higher than that in control group(P<0.05),and the level of survivin m RNAin CD4~+ T cells in nasal mucosa of SAR group was also higher than that in control group.After treatment with AICD activator,the frequency of apoptotic cells in the nasal mucosa of CD4~+ T cells in SAR group was significantly lower than that in the control group(P<0.05).The above results were statistically significant.Conclusion:Compared with the control group,survivin was highly expressed in the nasal mucosa CD4~+ T cells of SAR patients,and the frequency of CD4~+ T cell apoptosis was significantly decreased in nasal mucosa.Survivin may inhibit local CD4~+ T cell apoptosis.Participate in the formation of chronic inflammatory response states of allergic rhinitis. |
