Objective:In this work we investigate the effect of Yupingfeng on the MPI(minimal persistent inflammation)in allergic rhinitis and provide an experimental basis for further application of the Yupingfeng.Methods:Fifty female SPF BALB/c mice were divided into five groups randomly:low-dosage group,middle-dosage group,high-dosage group,Budesonide positive control group and MPI blank control group,ten mice for each group.All groups were injected intraperitoneally with the sensitizing solution containing OVA(ovalbumin)mixed with Al(OH)3 weekly for 3 weeks.And from day 22,they were all challenged intranasally daily with 40 mg OVA for one week.After the successful establishment of the AR model,from day 29,group A,B,C,D were challenged with 4 mg OVA mixed 4.88mg,9.75mg,19.5mg Yupingfeng and 0.33μg budesonide respectively for a long-term intranasal stimulation,lasting for three weeks.Group E were always conducted with 4 mg OVA in PBS solution for establishing MPI status without intervention.At last,the nasal symptom,the degree of nasal mucosal EOS infiltration and the expression of ICAM-1 in the epithelial cell were evaluated.Results:After nasal stimulation,the symptom score was more than 5 among all mice,so AR models were successfully established,and there were no significant differences between groups(P>0.05).From day 29,the nasal symptom such as sneezing,scratching and other symptoms disappeared gradually in group A,B,D and E,and significant differences from group C(P<0.01)were observed.The degree of nasal mucosal EOS infiltration in the epithelial cells in group A and B was lower than that of in group C and E(P<0.05),but greater than group D(P<0.05).Besides,there was no difference between group A and B(P<0.05).Group A and B had low expressions of ICAM-1 in the epithelial cells,whereas the expression in group C was high and group D had almost none expression.Furthermore,the group E(MPI blank control group)had a medium expression of ICAM-1 in the epithelial cells.Conclusions:By using the OVA,we can achieve the MPI status of allergic rhinitis model in BALB/c mice.Based on this,the low-dosage and the middle-dosage Yupingfeng can avoid coming up of the status of MPI in allergic rhinitis.It can providing an experimental basis for further study and good prevention of MPI of allergic rhinitis.By the way,the effect of Yupingfeng is lower than the Budesonide. |
