Objective To establish an animal model of ectopic splitting combined with portal vein ligation of rats and to explore the effect of that on rat liver regeneration.Methods Ten male SD rats weighing 220-250g were randomly divided into two groups(n=5):modeling group and control group.The control group groped for experimental conditions such as the dose of anesthetics,calculated the weight of each(sub)lobe of the liver.The modeling group used to establish an animal model of ectopic splitting combined with portal vein ligation(PVL),modelling of PVL(n=1),associating liver partition with PVL(ALPP)(n=2),associating spleen partition with PVL(ASPP)(n=1)and associating kidney partition with PVL(AKPP)(n=1).The right middle lobe of the liver was weighed on 10d postoperatively.The remaining 60 male SD rats were randomly divided into 4 groups(n=15):PVL group,ALPP group,ASPP group and AKPP group.Hepatic regeneration rate(HRR),ALT,AST and liver tissue proliferating cell nuclear antigen(PCNA)were detected on the 2nd,4th,and 7th day postoperatively.Results 2%,40mg/kg pentobarbital anesthesia was suitable,the liver accounted for3.08%of the body weight,the right middle lobe(RML)of the liver accounted for 0.74%of the body weight.Modeling success rate was 80%.In the modeling group,the RML weight of rats was5.33±0.4g,in the control group,the weight was 2.31±0.2g.After PVL combined with liver,spleen or kidney partition,the right middle lobe developed marked hyperplasia(F=17.41,P<0.01).On the 4th and 7th day,in ALPP group,ASPP group and AKPP group,there was an obvious hyperplasia(F=3.750,P<0.05;F=16.398,P<0.01)in the RML of the liver and higher PCNA(F=6.860,P<0.01;F=3.810,P<0.05)in the liver tissue than in PVL group.And ALPP group had significantly higher ALT and AST level on the 2nd and 4th day(ALT:F=8.812,F=21.929;AST:F=36.393,F=6.468;P<0.01).There was no significant difference in HRR or the positive expression number of PCNA in liver tissue among ALPP group,ASPP group and AKPP group.Conclusions We first successfully established a rat model of ectopic splitting combined with PVL.The success rate of modeling and postoperative survival was 80%.Ectopic splitting combined with PVL can significantly promote rat’s liver regeneration,possibly by inflammatory response,stress response and high expression of certain cytokines.
