| Panax quinquefolium L.(American ginseng,western ginseng)is a kind of ginseng plant belongs to the genus Araliaceae.It has a medicinal history for more than two-hundred year with the function of nourishing yin to reduce pathogenic fire,invigorating qi and promoting fluid metabolism.In recent years,with the development of Chinese medicine industry,a lot of effective research has been done on the chemical constituents and pharmacological effects of saponins,fatty acids,volatile oils,sugars and amino acids in American ginseng.In this paper,the chemical constituents of the new variety Zhongnong Yangshen No.1 were extracted and identified by medium pressure preparative chromatography,silica gel column chromatography,high performance liquid chromatography tandem mass spectrometry.At the same time,the discrepancies in saponin and sugar from different parts of Zhongnong Yangshen No.1,2 and four or five-year Zhongnong Yangshen No.1were compared,providing theoretical basis for the rational development,utilization and quality evaluation of the new variety Zhongnong Yangshen No.1.1.Extraction and separation of chemical components of Zhongnong Yangshen No.1The ginsenoside monomer in the new cultivar Zhongnong Yangshen No.1 was isolated and purified by medium pressure preparative chromatography combined with LC-MS.The result shows eight compounds were isolated and identified from the new variety Zhongnong Yangshen No.1,namely ginsenoside Re,ginsenoside Rg1,ginsenoside Rc,ginsenoside Rb1,ginsenoside Ro,ginsenoside Rb2,ginsenoside Rd,ginsenoside F11 with monomer purity of 98.2%,97.7%,93.2%,98.7%,94.8%,98.9%,96.9%,and 97.9%.2.Research on the difference of ginsenoside and sugar content in different parts of four or five-year Zhongnong Yangshen No.1(1)The difference of main ginsenosides content in different parts of four or five-year Zhongnong Yangshen No.1 were compared.The new method to measure the malonyl ginsenoside in western ginseng was established.The study found that the content of Re,Ro,Rb3 and Rd in the four-year-old Zhongnong Yangshen No.1 was significantly higher than that in five-year-old in the basal part of stem.In tassel,the content of Rb1,Rc and Rb3 in the four-year-old Zhongnong Yangshen No.1 was significantly lower than that in the five-year-old Zhongnong Yangshen No.1,and there were no significant difference between the xylem and the phloem.Analysing the malonyl ginsenoside,in the basal part of stem,the content of M-Rb3 and M-Rd in the four-year Zhongnong Yangshen No.1 was significantly lower than that in the five-year-old Zhongnong Yangshen No.1,and there were no significant difference between the xylem and the phloem.(2)Compaing the difference of sugar content in different parts of four or five-year Zhongnong Yangshen No.1.The study found that the content of fructose and glucose in the four-year-old ginseng ginseng No.1 was significantly higher than that in five-year Zhongnong Yangshen No.1 in the basal part of stem;In phloem,the contents of glucose,sucrose and maltose in four-year Zhongnong Yangshen No.1were significantly higher than that in five-year Zhongnong Yangshen No.1;In tassel,the content of fructose and sucrose in four-year Zhongnong Yangshen No.1were significantly higher than that in five-year Zhongnong Yangshen No.1,and the glucose content was significant lower than that in five-year Zhongnong Yangshen No.1.3.Discrepancies in different parts Zhongnong Yangshen No.1 and Zhongnong Yangshen No.2 of saponins and sugar contents.(1)The content of main ginsenosides in different parts of Zhongnong Yangshen No.1 and Zhongnong Yangshen No.2 was determined by ultra-high pressure liquid chromatography(UPLC).In root,the Re,Ro,Rb1 contents in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2.In tassel,the Re,Ro,Rb3 contents in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2.Analysing the malonyl ginsenoside,we found that in the basal part of stem,the M-Rc,M-Rb3 and M-Rd contents in Zhongnong Yangshen No.1 was significantly higher than that in Zhongnong Yangshen No.2.In root,the M-Rc content in Zhongnong Yangshen No.1 was significantly higher than that in Zhongnong Yangshen No.2.In tassel,the M-Rb1 content in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2.(2)The difference in main sugar including glucose,fructose,sucrose and maltose contents between different parts of Zhongnong Yangshen No.1 and Zhongnong Yangshen No.2 was determined by UPLC method.The results showed in the basal part of stem,the sucrose content in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2.In root,the sucrose and maltose contents in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2,but the fructose was higher.In tassel,the fructose and sucrose contents in Zhongnong Yangshen No.1 was significantly lower than that in Zhongnong Yangshen No.2,but the glucose was absolutely higher. |
