Objective(s):By detecting the expression levels of long non-coding RNA(lncRNA)Metastasis-associated lung adenocarcinoma transcript 1(MALAT1)to analysis expression changes and significance of lncRNA MALAT1 in high risk population of non-alcoholic steatohepatitis(NASH).So we can look for a possible diagnostic marker for early identification in NASH high-risk populations.Methods:A total of 95 patients with non-alcoholic fatty liver disease(NAFLD)and 46 patients without NAFLD were randomly selected for anthropometric and metabolic indicators.The total RNA of peripheral blood mononuclear cells was extracted,and the expression level of lncRNA MALAT1 in peripheral blood of each group was detected by qRT-PCR.ELISA was used to detect the serum levels of CK-18 in the NAFLD group.According to the 2018 version of the guidelines for the prevention and treatment of nonalcoholic fatty liver disease,the NAFLD component was recommended to be a simple NAFLD group(n=55 cases)and a NASH high risk group(n=40 cases).The expression levels of lncRNA MALAT1 in the non-NAFLD group,the simple NAFLD group and the NASH high-risk group were compared.Results:1.Compared with the non-NAFLD group,the body weight,BMI,waist circumference,waist-to-hip ratio,UA,TG were significantly increased in the NAFLD group,and HDL-c was significantly decreased.There was no significant difference between the simple NAFLD group and the NASH high-risk group.2.The relative expression of lncRNA MALAT1 in the non-NAFLD group,simple NAFLD group and NASH high risk group was statistically different(P<0.05).Compared with the non-NAFLD group,the expression level of lncRNA MALAT1 was significantly increased in the high-risk group of NASH(P=0.005).The expression level of lncRNA MALAT1 in the high-risk group of NASH was significantly higher than that in the simple NAFLD group(P=0.002).The expression level of lncRNA MALAT1 was not significantly different between the non-NAFLD group and the simple NAFLD group(P>0.05).Conclusion(s):1.Compared with the non-NAFLD group,patients with NAFLD have overweight and abdominal obesity;they are accompanied by metabolic abnormalities of uric acid and lipid profiles.2.The relative expression level of lncRNA MALAT1 was significantly elevated in the high-risk population of NASH.It is speculated that lncRNA MALAT1 may be one of the indicators for assessing the risk of developing NASH in patients with NAFLD.
