Objection: Quantitative analysis of the expression of inducible nitric oxide synthase(i NOS)in the maxillary sinus mucosa of patients with maxillary sinus fungus ball sinusitis and chronic sinusitis without nasal polyps by Western blotting technique,to explore i NOS in maxillary sinus fungus ball sinusitis and chronic sinusitis without nasal polyps in patients with maxillary sinus mucosa expression difference and its correlation with maxillary sinus fungus ball sinusitis and chronic sinusitis without nasal polyps.Methods: A total of 51 patients who underwent functional endoscopic sinus surgery under general anesthesia in the Department of Otorhinolaryngology of the Second Affiliated Hospital of Dalian Medical University from October 2017 to October 2019 were collected.All patients were divided into 3 groups,17 cases in each group,namely chronic sinusitis without nasal polyps group,maxillary sinus fungus ball sinusitis group,normal control group(simple maxillary sinus cyst).All patients were excluded from bronchial asthma,allergic rhinitis,autoimmune diseases,ciliary syndrome and cystic fibrosis,and the maxillary sinus cyst group was excluded from the history of acute and chronic nasal inflammatory diseases.During general anesthesia,the remaining medial maxillary sinus mucosa of the three groups except the pathological specimens were collected,and the weight of each mucosal tissue in each group was no less than 30 mg.The total protein was extracted from the mucosal tissues,and the protein content in the supernatant was detected by Western blot by electrophoresis,electric transfer,sealing,incubation of the primary antibody,incubation of the secondary antibody and development,and the corresponding Western blot map was obtained.The primary antibody of the Western blot was anti-rabbit i NOS,and the secondary antibody was goat anti-rabbit antibody.The experiment results were imported by Image studio to analyze the gray value and corresponding ratio of each strip,and then Graphpad statistical software was used to plot and analyze the differences among the three groups.Spearman correlation was used to analyze the correlation between i NOS and maxillary sinus fungus ball sinusitis and chronic sinusitis without nasal polyps.Test standard: P <0.05 is considered to be different,P <0.01 is considered to be significant;the absolute value of r determines the degree of correlation.Results: 1.The expressions of i NOS in the maxillary sinus mucosa of maxillary sinus fungus ball sinusitis group and chronic sinusitis without nasal polyps group were higher than those of the simple maxillary sinus cyst group,with statistically significant differences(P<0.05);2.i NOS is highly expressed in the maxillary sinus mucosa of the maxillary sinus fungus ball sinusitis group,which is significantly different from the chronic sinusitis without nasal polyps group in maxillary sinus(P<0.01);3.The expression of i NOS was positively correlated with the incidence of maxillary sinus fungus ball sinusitis(r=0.6395,P<0.05),while was weakly correlated with the incidence of chronic sinusitis without nasal polyps(r=0.3396,P<0.05).Conclusion: 1.i NOS is positively correlated with the incidence of maxillary sinus fungus ball sinusitis,however the incidence of chronic sinusitis without nasal polyps is weakly correlated.INOS plays an important role in the pathogenesis of maxillary sinus fungus ball sinusitis.2.The quantitative detection of i NOS in the sinus mucosa can indirectly and accurately reflect the level of NO in the sinus.
