Part one Effects of Silencing SOX21-AS1 on Proliferation,Invasion and Migration of Cervical CancerObjective:To investigate the effect of long-chain non-coding NRA SOX21-AS1(lncRNA SOX21-AS1)on the proliferation,invasion and migration of cervical cancer HeLa cells.Methods:Cervical cancer HeLa cell lines were divided into three groups.The interference SOX21-AS1(sh-SOX21-AS1)group was transfected with sh-SOX21-AS1,and the interference control(sh-NC)group was transfected with sh-NCand untreated cells was blank group.CCK-8 assay was applied to examine cell proliferation activity.Transwell invasion test was used to detect cell invasion ability,and Transwell method was used to detect cell migration ability.Results:After the expression of lncRNA SOX21-AS1 was silenced,the CCK-8 assay showed that the absorbance value of sh-SOX21-AS1 group was(0.77±0.56),which significantly increased compared with that of the blank group(0.54±0.01)and sh-NC group(0.59±0.22)(P<0.05).The results of the Transwell invasion experiment showed that the relative cell number of the cells in the sh-SOX21-AS1 group was(109.67±11.59),whichsignificantly increased compared with the blank group(78.67±6.44)and the sh-NC group(88.33±5.17)(P<0.05).Transwell assay to test cell migration ability after silencing lncRNA SOX21-AS1 showed that the number of migrating cells in the sh-SOX21-AS1 group(106.00±9.00)significantly increased compared to sh-NC(81.00±10.14)and blank group(82.33±10.01)(P<0.05).Conclusion:1.Silence of lncRNA SOX21-AS 1 promotes proliferation of cervical cancer cells.2.Silence of lncRNA SOX21-AS1 promotes the invasion of cervical cancer cells.3.Silence of lncRNA SOX21-AS1 promotes migration of cervical cancer cells.Part two the Relationship between NLR and PLR and clinical characteristics of cervical cancerObjective:To explore the correlation between serum neutrophil-lymphocyte ratio(NLR),platelet-to-lymphocyte ratio(PLR)and clinical characteristics of patients with cervical cancer.Methods:A retrospective analysis of 209 patients with cervical cancer treated for the first time in the gynecology or oncology department of the Affiliated Hospital of North Sichuan Medical College(hereinafter referred to as "our hospital")from January 2016 to December 2017 was the study object,with a median age of 50 years(23~76 years old),and collected 200 healthy women who were examined at the health management center of our hospital during the same period as the control group,with a median age of 51 years(23~74 years).The differences between NLR and PLR in the two groups were analyzed.The correlation between NLR and PLR in patients with cervical cancer was analyzed.To analyze the correlation between NLR and age,FIGO stage,tumor size,pathological type,pathological grade,lymph node metastasis and Lymphovascular Space Invasion in patients with cervical cancer.To analyze the correlation between PLR and age,FIGO stage,tumor size,pathological type,pathological grade,lymph node metastasis and Lymphovascular Space Invasion in patients with cervical cancer.Results:The cervical cancer group NLR(2.89±1.86)was significantly higher than the control group NLR(1.85±0.52)(P<.001).Cervical cancer group PLR(147.93±84.35)was significantly higher than control group PLR(110.05±30.08)(P<0.001).Patients with elevated NLR(NLR≥2.39)had worse pathological differentiation,larger tumor size,later FIGO stage,more lymph node metastases and more Lymphovascular Space Invasion than patients with reduced NLR(NLR<2.39)(P<0.05).Patients with elevated PLR(PLR≥120.98)had more lymph node metastasis than patients with reduced PLR(PLR<120.98)(P<0.05).PLR is not related to tumor size,FIGO stage,Lymphovascular Space Invasion,age,pathological grade and pathological type(P>0.05).There was a significant positive correlation between NLR and PLR(r=0.743,P<0.05).Conclusion:NLR and PLR were elevated in peripheral blood of patients with cervical cancer in our hospital,and NLR and PLR had a significant positive correlation.The elevation of peripheral blood NLR in patients with cervical cancer is related to FIGO stage,tumor size,lymph node metastasis,Lymphovascular Space Invasion and pathological grade.It has certain reference value for the prognosis of patients with cervical cancer.PLR only has relationship with lymph node metastasis in patients with cervical cancer,but has nothing to do with FIGO stage,tumor size,Lymphovascular Space Invasion,age,pathological type,and pathological grade.It has limited predictive value for patients with cervical cancer.
