Objective:To investigate the relationship between Hcy and MTHFR C677T gene polymorphism and coronary heart disease in eastern yunnan provinceMethods:Patients admitted to the department of cardiology,qujing first people's hospital from June 2018 to July 2019 were selected for coronary angiography or coronary CTA examination.The results indicated that 212 patients were diagnosed with coronary heart disease,including 140 males and 72 females,with an average age of 61.94 ± 10.96 years.210 patients without coronary heart disease were selected as the control group,including 85 males and 125 females,with an average age of 53.07±14.53 years.The admission number,name,sex,age,history of hypertension,history of diabetes,serum homocysteine,MTHFRC677T genotype,blood lipid(including cholesterol,triglyceride,HDL,LDL)of all the selected subjects were collected.Genotype analysis was performed using PCR-restriction fragment polymorphism.Plasma Hcy concentration was determined by enzyme cycle rate method.The differences in Hcy level and MTHFR genotype between the CHD group and the control group were compared,and the allele C and allele T of the two groups were calculated respectively to compare whether the allele T increased the risk of CHD.The subjects were divided into groups at the age of 60 to compare whether there were differences in MTHFR genotypes between the CHD group at the age of 60 and the control group,and whether there were differences in MTHFR genotypes between the CHD group at the age of 60 and the control group.According to the subtypes of coronary heart disease,the coronary heart disease group was further divided into the stable angina pectoris group and the ACS group,and the differences of Hcy level and MTHFR genotype between the two groups were respectively compared.Among all the included researchers,Hcy levels of genotype CT and TT were compared with genotype CC to explore the relationship between MTHFR genotype and Hcy.All experimental data were statistically analyzed using SPSS 25.0.The measurement data were expressed as mean ± standard deviation(x ± s),According to the results of homogeneity test of variance,if it conforms to the normal distribution,the independent sample T test is adopted;if it does not conform to the normal distribution,the rank sum test is adopted..The enumeration data were expressed by the composition ratio or rate,and the comparison between groups was?2tested.The difference of P<0.05 was statistically significant,and the influence of confounding factors was adjusted by logistic regression analysis.Results:1.The Hcy level was 15.9±9.45umol/L in the CHD group and 11.89±5.94umol/L in the control group.P<0.05),the Hcy level of CHD group was higher than that of the control group,and the difference was statistically significant(P<0.05).2.There was no significant difference in MTHFR genotype between the CHD group and the control group(P>0.05).3.There was no significant difference in allele C and allele T between the CHD group and the control group(P>0.05).4.There was no significant difference in MTHFR genotype between the CHD group and the control group in the over-60s and the under-60s(P>0.05).5.There was no significant difference in MTHFR genotype among women(P>0.05).There was no significant difference in MTHFR genotype among men(P>0.05).6.There was no significant difference in blood homocysteine levels and MTHFR genotype between the acute coronary syndrome group and the stable angina group(P>0.05).7.With the same genotype at MTHFR 677,The Hcy level in the CHD group was significantly higher than that in the CHD group,(Hcy:C/Ctype:CHD14.11±9.02umol/L,control group,11.78±8.03umol/L;C/T type:CHD14.11 ±5.56umol/L?control group 11.98±4.65umol/L,P<0.05;T/T type:CHD26.00±14.19umol/L,control group 11.88±3.31umol/L,P<0.05).8.To compare the standard with wild type C/C type,there was no significant difference in blood Hcy level in C/T group(P>0.05),while the blood Hcy level in T/T group was significantly increased,The difference was statistically significant(P<0.05).(Hcy:C/C:14.1 1±9.02umol/L,C/T:14.1 1±5.57umol/L,T/T:26.00±14.19umol/L)9.In all patients with coronary heart disease(CHD),to compare the standard with wild type C/C type,there was no significant difference in blood Hcy level in C/T group(P>0.05),while the blood Hcy level in T/T group was significantly increased,The difference was statistically significant.(P<0.05).(Hcy:C/C:14.11 ±9.02umol/L,C/T:14.11±5.57umol/L,T/T:26.00±14.19umol/L)?Conclusion(s):1.Homocysteine was a high risk factor for coronary heart disease.2.there was no significant correlation between MTHFR gene polymorphism and CHD.Allele T was not associated with increased risk of coronary heart disease in eastern Yunnan.3.The homocysteine level of MTHFR677 genotype T/T was significantly higher than that of C/C,C/T genotype in eastern Yunnan.4.In patients with coronary heart disease,the homocysteine level of MTHFR677 genotype T/T was significantly higher than that of C/C and C/T genotype in eastern Yunnan.
