| Objectives: Through the early intervention of Yangxinxue granule on chronic heart failure model rats induced by doxorubicin,the effects of Yangxinxue granule on general condition,cardiac color ultrasound,whole heart mass index,cardiological changes and ATP content in myocardial tissue were observed to evaluate the preventive therapeutic effect of Yangxinxue granule on chronic heart failure model rats,and to explore the mechanism of Yangxinxue granule in protecting myocardium and cardiac function.Experimental methods: The model of chronic heart failure was induced by intraperitoneal injection of doxorubicin in SD rats.2mg/kg was injected intraperitoneally according to the body weight of rats,twice a week in the first 2 weeks and once a week in the last 6 weeks to the cumulative dose of 20mg/kg.80 male SD healthy rats were randomly divided into 6 groups: blank group,traditional Chinese medicine control group,model control group,low-dose Yangxinxue granule group,middle-dose Yangxinxue granule group and high-dose Yangxinxue granule group.The model control group and Yangxinxue granule high,middle and low dose groups were intraperitoneally injected with doxorubicin,while the blank group and traditional Chinese medicine control group did not make the model.One day before the establishment of the model,the rats in the traditional Chinese medicine intervention group were fed with the corresponding dose of Yangxinxue granule,the traditional Chinese medicine control group was fed with high-dose Yangxinxue granule,and the model group and blank control group were given equal volume of drinking water.And continuous intragastric administration of Yangxinxue granule in the process of modeling in order to maintain the stability of its blood concentration.After 8 weeks of modeling,the cardiac function of rats was evaluated by echocardiography,heart samples were collected,myocardial HE staining and Masson staining were performed to observe the pathological changes of rat heart tissue,and the content of AT P in myocardial tissue was detected to evaluate myocardial energy metabolism.Results: 1)General observation of rats: the model group and each dose group of Yangxinxue granule showed different degrees of heart failure,including deterioration of mental state,decrease of activity and food intake,dry hair,abdominal swelling and so on.The body weight and heart weight were significantly lower than those in the blank group and the traditional Chinese medicine control group.(P<0.01).2)it was confirmed that the model was successful: eight weeks after the establishment of adriamycin model,the levels of EF,SV,CO,IVSs and LVPWs in the model group were significantly lower than those in the blank group(P<0.01).3)the results of echocardiography of rat heart: the measured values of EF,FS,IVSs and LVPWs in each group of Yangxinxue granule were higher than those in the model group,but there was no significant difference between the traditional Chinese medicine control group and the blank group.4)the content of ATP in myocardial tissue of model group was significantly lower than that of blank group,traditional Chinese medicine control group and Yangxinxue granule groups(P<0.01).5)Under HE staining,the transverse striations of cardiomyocytes in the blank group and the traditional Chinese medicine control group were clear,and there was no obvious angiogenesis and inflammatory cell infiltration.In the model group,the staining of cardiomyocytes was uneven,the transverse lines were blurred,the volume of some cells decreased,the plasma staining deepened,and the number of small blood vessels between myocardium increased;in the low,middle and high dose groups of Yangxinxue granule,the transverse lines of cardiomyocytes were clear and the number of blood vessels between myocardium increased.Under Masson staining,the muscle fibers in the blank group were evenly distributed,the collagen fibers around the myocardium were sparse,and the fiber width was thinner.The proliferation of distal fibers around the muscle fibers was not obvious in the low,middle and high dose groups,which was similar to that in the blank group.In the model group,the staining of muscle fibers was uneven,light blue collagen could be seen in some cardiomyocytes,the number of collagen fibers around cardiomyocytes increased and the width of collagen fibers widened.Conclusion: 1)the rat model of chronic heart failure induced by intraperitoneal injection of adriamycin was successfully established.2)Early intervention of Yangxinxue granule on rats with chronic heart failure induced by adriamycin can effectively improve cardiac function,protect mitochondrial function,improve myocardial energy metabolism,prevent myocardial injury and fibrosis,and prevent and slow down the occurrence of chronic heart failure. |
