Among the common gynecological malignancies in female reproductive organs,the incidence of ovarian cancer is second only to cervical and endometrial cancer,and the mortality rate is the first.Because it often has no obvious symptoms in the early stage,most of the findings are late,and the degree of malignancy is high,it can be transferred through abdominal implantation,local diffusion,lymphatic metastasis and blood metastasis.At present,the main principle of ovarian cancer treatment is surgical adjuvant chemotherapy.Surgery and chemotherapy work well for most early patients,but little for most late patients.The big data show that the incidence of ovarian cancer in developed and developing countries is 91/100,000 and 50/100,000,respectively[1].due to the high resistance of platinum and yew classes,more than 25%of advanced patients developed recurrence 6 months after starting treatment[2-3].With the development of advanced disease and chemotherapy resistance,the average survival rate of most patients is only 4.5 years.Although many drugs have been developed to target ovarian cancer,they are expensive and have many side effects.generally speaking,the ideal targeted anti-tumor drug can selectively kill tumor cells,but the current targeted drug used for anti-tumor can kill tumor cells at the same time,it will also cause accidental injury to the surrounding normal cells,and produce some toxic and side effects on the human body.In view of the present form,seek a security A fully effective treatment for ovarian cancer is particularly necessary.Dithiolam?Disulfiram,DSF?is an alcohol abstinence drug.It has been used in clinical practice for more than 70 years because of its low price,low toxicity and good safety.the study found that dithiolam can efficiently kill tumor cells without affecting the surrounding normal cells[4]].Therefore,scientists boldly speculate that dithiolam has anti-tumor effect.DSF are common sulfur-containing ligands that can form stable complexes with most metals.Copper is one of the essential trace elements of human body and is indispensable to maintain the normal life activities of organisms.It was found that copper ions in tumor cells were significantly higher than those in normal cells[5-6].After a large number of research scientists further found:DSF can and copper ions to form a stable induction of apoptosis of the complex[7].Objective:Through in vitro cell experiments,copper?copper,Cu?was given a certain dose alone,different doses of DSF and a certain dose of Cu were combined with different doses to observe the changes of cell morphology and number SKOV3 human ovarian cells,and to explore the effect of DSF/Cu on the inhibition of proliferation and induction of cell apoptosis in human ovarian cancer cells.Methods:1.Cell culture:Human ovarian cancer SKOV3 cell line was cultured in DMEM/H medium.The conditions were 37°C and 5%CO2.2.Experimental grouping:This experiment is divided into a blank control group,a 0.5?mol/LCu group,a concentration of 0.2?mol/L,0.4?mol/L,o.6?mol/L,0.8?mol/L DSF,and a combination of DSF at the above concentrations Cu treatment group3.Thiazole blue method?MTT?to detect the inhibition of cell proliferation after drug intervention for 48 hours4.Flow cytometry to detect apoptosis rate5.Statistical method:SPSS22.0 statistical software was used to process the experimental data.P<0.05 was considered statistically significant.Result:1.The results of MTT showed that DSF and DSF/Cu inhibited the proliferation of human ovarian cancer cell SKOV3 and were affected by the concentration of DSF?P<0.05?.Among them,DSF/Cu had a significantly stronger inhibitory effect on cell proliferation than DSF alone.The difference was statistically significant?P<0.05?.There was no significant difference in the inhibition rate of cell proliferation between the Cu group and the blank group?P>0.05?.2.Flow cytometry showed that 0.4?umo L/L of DSF combined with Cu for 48 hours compared with the blank group,the apoptosis rate in the DSF/Cu group was significantly higher than that in the blank group,and there was no statistical difference between the Cu group and the blank group.Significance?P>0.05?.Conclusions:1.Both DSF and DSF/Cu can inhibit the proliferation of human ovarian cancer SKOV3 cells.DSF combined with Cu has a significantly stronger effect than DSF alone.2.DSF/Cu can induce apoptosis of human ovarian cancer cell SKOV3.
