Objective:To study the chemical constituents from the dried rhizome of Menispermum dauricum DC.,especially the alkaloids.To study the absolute configurations of the abietanes from Euphorbia fischeriana Steud.Methods:1.The dried rhizome of Menispermun dauricum was firstly extracted with 95% ethanol by reflux.The combined extracts were evaporated under reduced pressure and the residue was dissolved in aqueous acid.The acidic solution was made alkaline and extracted exhaustively with organic solvent for total alkaloids.The total alkaloids were separated and purified by various methods to obtain monomers.Their structures were identified by spectral analysis and comparison with physical and chemical properties.2.The 95% ethanol extract from the dried roots of Euphorbia fischeriana Steud was extracted by ethyl acetate.Four abietane-type diterpenes were isolated from the ethyl acetate extract and were successfully identified on basis of the spectroscopic means.The absolute configurations of 17-Hydroxyjolkinolide A(1)and 17-Hydroxyjolkinolide B(2)were determined by X-ray diffraction,CD,ECD and other methods.Results:In this research,ten alkaloid compounds were isolated and identified from Menispermum dauricum as: daurisoline(1),sinomenine(2),armepavine(3),O-methylcorypalline(4),pecrassipine B(5),dauricine(6),corypalline(7),(R)-6-methoxy-1-(4-methoxybenzyl)-2-methyl-1,2,3,4-tetrahydroisoquinolin-7-ol(8),dauricumidine(9),O-methyldauricine(10);The absolute configuration of 17-Hydroxyjolkinolide A(1)was determined as(5R,8S,9S,10 R,14R)and 17-Hydroxyjolkinolide B(2)was determined as(5R,8S,9R,10 R,11R,12 R,14R)from Euphorbia fischeriana.Conclusion:1.Among the ten alkaloids isolated,compounds 1,6 and 10 are alkaloids with dibenzyl isoquinoline structure,compounds 3,5 and 8 are alkaloids with monobenzyl isoquinoline structure,compounds 2 and 9 are alkaloids with morphine-type structure,and compounds 4 and 7 are alkaloids with tetrahydroisoquinoline structure.In addition,compounds 3,5 and 8 were isolated from this plant for the first time.2.In this paper,the relationship between the ECD spectra and the absolute configuration of the butenolide of abietane on basis of the X-ray diffraction result was studied,which make it more accurate to determine the absolute configuration using ECD.It can provide reference and criterion for the determination of the absolute configuration of 12 carbon in butanolide of abietane.
