[Objective] To investigate the effects of antitumor and oxidation resistance ability in H22tumor-bearing mice by LDE extracted from Euphorbia Fisheriana Steud.[Methods] With H22tumor-bearing mice as the research object in this study, the trials are divided into two parts, that is, inhibitory tumor experiment and antioxidant experiment, respectively. Inhibitory tumor experiment mainly includes:to calculate the inhibitory rate; to calculate apoptosis ratio by Flow Cytometry; to observe the morphological changes of apoptosis by labeling with fluorescence probe analysis(Annexin-V FITC);to observe the morphological changes of apoptosis by transmission electron microscope; to observe the effects of living conditions of mouse after treatment. Antioxidant experiment mainly includes:to analysize the liver index and the spleen index, to detect the vitality of the value of CAT, SOD and GSH-PX in mice serum by754Spectrophotometer.[Results]1. The measurements of the tumor-bearing mice after14days treatment shows that the LDE has significantly inhibited the H22tumor growth, and when LDE combined with microwave hyperthermia, its inhibitory effect is shown to be more apparent.2. By Annexin-V FITC/PI double labeling staining results shows that, the LDE can clearly induce apoptosis, and when it combined with microwave thermotherapy, the promoting effect of H22cell apoptosis is more evident. By immunofluorescence microscope, LDE group and LDE combined with microwave thermotherapy group are concluded that there are a large number of Annexin-V FITC and PI staining cells, indicating that there is a large number of apoptosis occurring in H22cells.3. Observed by TEM, LDE group and The LDE combined with hyperthermia group appeared the typical change of apoptosis, the Endoplasmic Reticulum and Golgi complex are swelling, vacuated, the Mitochondrial crista blured, or even disappeared; the pronuclear space is expanding, varying sizes, the nuclear fragmented and the nuclear shrinkaged, the tumor cell membrane budding, displaying apoptotic bodies in late phase of apoptosis.5. LDE can enhance the liver index and spleen index in the H22-bearing mice, and when it combined with microwave thermotherapy, the effect is more obvious.6. LDE can induce the serum levels of SOD, GSH-PX and CAT in H22-transplanted mice, therefore,enhancing the antioxidant capacity of tumor-bearing mice. And when LDE combined with microwave thermotherapy, the effect is more obvious.[Conclusion]LDE is an effective component against proliferation of malignant H22Hepatocarcinoma cells. The mechanisms of action for LDE against H22Hepatocarcinoma cells may be the induction of the apoptosis as well as suppression of angiogenesis by affecting indexes such as the relevant SOD, GSH-PX or CAT activity in the cancer cells.1. LDE has showed remarkably antitumor effect, and more medicinal safety, the water-soluble extract of Lang-Du has stronger inhibition effect when it combined with microwave hyperemia than when used by each one alone.2. The water-soluble extract of Lang-Du has strong antioxidant capacity and it has stronger effect when it combined with microwave thermo-hyperemia.
