| Purpose:1.To provide scientific experimental data for identificating the Cinnamomum plant ’by biology research and the chemical study.2.Looking for compounds with antitumor activity to provide a scientific basis for comprehensive exploitation and utilization.Methods:1.Identificology research:using method of morphological Characteristics and microscopic identification to study the biological characteristics of the leaves of CCCB;2.Analysis of the volatile oil:volatile oil were extracted by the steam distillation method,and the chemical composition of leaf essential oil were analyzed by GC-MS;3.Study of chemical compositions of EtOAc:Silica gel,HP-20,Sephadex LH-20,RP-18,PTLC were used to isloate compounds.Their stuctures were identificated by spectrum analytical method(UV,IR,1H-NMR,13C-NMR,DEPT,HSQC,HMBC,1H-1HCOSY,NOESY,MS).4.Screening compounds with antitumor activity:reduce rate of NADPH within 10 min was evaluated by continuous monitoring method to determine initial velocity of AKR1C1/C2/C3 catalyzing reduction 9,10-phenanthrenequinone,then calculating inhibition rate.Results:1.The characteristics of identificology:The leaves of CCCB are broadly ovoid,leaf apex acutus;the surface of Petiole is rough,furcella occasionally;Gland is big and deep,expanded at leaf vein;unique strong fragrance.Leaf transverse sections:on main veins skin for a list of long flat square cells;Outside the main vein vascular bundle in the infinite toughening type,distribution of fiber bundles around the vascular bundles;Palisade cell 2 columns,spongy tissue rich oil cells;Under the epidermis stomatal assumes the infinitive,non-glandular hairs were found accidentally.2.Analysis of the volatile oil:34 compounds have been found from the essential oil of the leaves of CCCB.Its main compositions are d-Borneol(49.76%),d-2-Bornanone(14.30%).And the content of 11 components is higher than 1%.3.Study of nonvolatile compounds:16compounds were isolated from CCCB,including 9 Phenylpropanoids,3 Flavonoids,2 Triterpenoids,1 fatty acid,1 salicylic acid.They aresesamin(1),8(R)-methoxy sesamin(2),8(S)-methoxy episesamin(3),8(R)-hydroxy sesamin(4a),8(R)-hydroxyepisesamin(4b),(1 R,2S,5 S,6S)-2-(3,4-dioxymethylenephenyl)-6-(4-hydroxy-3-methoxyph enyl)-8S-hydroxy-3,7-dioxabicyclo[3.3.0]octane(5a),(1 S,2S,5R,6R)-2-(3,4-dioxymethy-lenephenyl)-6-(4-hydroxy-3-methoxyphenyl)-8R-hydroxy-3,7-dioxabicyclo[3.3.0]octane(5b),Syringaresinol(6),Medioresinol(7),quercetin(8),kaempferol(9),5,7,4’-3 hydroxy-dihydro flavonoid(10),betulinic acid(11),[P-sitosterol(12),palmitic acid(13),Salicylic acid(14)respectively.4.Screening compounds with antitumor activity:inhibition rate to AKRlC(C3/C2/C1)of compound 1(sesamin)were 0%,4%,0%respectively;Compound 4(8-hydroxy sesamin)were 0%,9%and 0%respectively;Compound 10(5,7,4’-3 hydroxy-dihydro flavonoid)were 88%,69%,95%respectively;d-boneol crystallization were 0%,0%,30%;leaf essential oil were 0%,29%,3%.Conclusion:1.Compound 10(5,7,4’-3 hydroxy-dihydro flavonoid)showed strong inhibitory activity of AKR1C(C3/C2/C1);2.Compound 1~7 are the double tetrahydrofuran lignans,which are the skeleton feature of CCCB.This skeleton compounds were found from Cinnamomum plant for the first time.4.The other compounds were found from this plant for the first time. |
