An Experimental Study On The Intervention Of Zhengshi Qubi Compound On Early Ankylosing Spondylitis In Balb/c Mice

Objective: To study the characteristics and understanding of Prof essor Zheng Huaixian’s treatment of ankylosing spondylitis(AS)by studying the experience of Professor Zheng Huaixian,and to study the treatment of Balb/c mi ce with Zheng’s Chinese herbal compound.Early symptomology,histomorphology,and tumor necrosis factor-α(TNF-α),interleukin-1(IL-1β),and interleukinin in ankylosing spondylitis-2,IL-2),interleukin-6(IL-6),monocyte chemotactic protei n-1(MCP-1)concentration changes,explore its association with AS The early tre atment effect and possible mechanism will further improve Professor Zheng Huai xian’s traumatic prescription system and lay a solid foundation for its clinical ap plication.Methods Eighty female 10-week-old Balb/c mice were randomly divided into groups(n=20),model group(n=20),Chinese medicine fumigation group(n=20),argon nitrogen.Sulfasalazine(SSZ)and meloxicam combined with Western medi cine group(n=20),except for the blank group,the other three groups of mice w ere treated with complete Freund’s adjuvant plus proteoglycan(adjuvant 0.1ml + protein aggregation)Sugar 0.1ml)was intraperitoneally injected on the 0th,21 st,and 42 th day to remove the dead and modeled mice,set blank group(n=15),m odel group(n=15),Chinese medicine fumigation group(n= 15),Western medicin e group(n=15),the second day after the last intraperitoneal injection was compl eted.The Chinese medicine fumigation group was externally smoked and washed with Zheng’s Chinese medicine compound once every other day.The Western m edicine group referred to the "Pharmacological Experiment Methodology" and dil uted with a proper amount of physiological saline to make a turbid suspension t o the mice,once a day.The joint circumference was measured to observe the s welling of the joints of the mice.The changes of inflammatory factors such as TNF-α,IL-1β,IL-2,IL-6 and MCP-1 were evaluated by tissue section.Results: 1 Theoretical research results: Zheng’s sputum Chinese herbal comp ound is used to treat early AS with phlegm and collaterals,promoting blood circ ulation and relieving phlegm,and relieving pain and relieving pain.2 Results of animal experiment: The changes of laboratory indexes were significantly differen t between the model group and the serum group(P<0.01).The traditional Chines e medicine fumigation group had significant difference compared with the serum group of the model group(P<0.01),the Western medicine oral group was signifi cantly different from the model group(P<0.01).There was no significant differen ce in joint dimension between the groups before the start of the experiment.Onthe third day of the experiment,the model group,the Chinese medicine fumigat ion group and the western medicine group showed different degrees of external j oint swelling,the 28 th day.The swelling reached the peak.After the end of the experiment,there was a significant difference in the joint circumference betweenthe model group and the blank group(P<0.01).There was a statistically signifi cant difference in the joint circumference between the Chinese medicine fumigati on group and the model group(P<0.01).There was a statistically significant diff erence in joint circumference between the oral group and the model group(P<0.01).Histomorphological changes were observed.After the treatment of the smoke d drug group,the histological examination of the ankle joint and the cerebral pal sy was significantly changed in the model group.After the treatment of the west ern medicine group,the histological examination of the ankle joint and the cereb ral palsy was significantly changed.The model group was compared with the bla nk group,and the ankle joint and the sacral tissue were significantly changed.Conclusion: The compound prescription of Zheng’s sputum is based on phle gm and blood stasis,promoting blood circulation and relieving phlegm,relieving pain and relieving pain,and supplementing the liver and kidney for early treat ment of AS,and cutting the pathogenesis.Animal experiments show that the tre atment of early AS with Zheng’s Chinese herbal compound can interfere with th e treatment of early mandatory spondylitis by regulating inflammatory reaction,i mprove joint swelling and improve tissue morphology.
