| Purpose:In this experiment,the DOR rat model induced by cyclophosphamide was used as the carrier,the traditional Chinese medicine compound and the acupuncture and moxibustion prescription,which were used as the guiding ideology,were used as the intervention factors to observe the protein expression of factors related to apoptosis of ovarian granulosa cells,such as Bcl-2,Bax and p53,and to explore the effect of Tongbu Qijing therapy on the ovarian granulosa cells of DOR rats as well as it’s mechanism of action.Material and method:Forty-five SD female rats were randomly divided into normal group,model group,traditional Chinese medicine group,acupuncture group and acupuncture group according to random number table.In addition to the normal group,other groups were given intraperitoneal injection of cyclophosphamide(50mg / kg / d)a week,DOR modeling,vaginal smears observed estrous cycle,the emergence of a period of extension or disappearance of modeling success.The successful model rats were treated with traditional Chinese medicine gavage,acupuncture,gavage and acupuncture intervention method,the intragastric dose of 3mg / kg / d,continuous gavage for 4 weeks;acupuncture use Huatuo licensing 0.18 × 13 mm Acupuncture needles,acupoints for the Guanyuan,Baihui,Mingmen,Gongsun,Zhaohai,day 1,needle 20 min,acupuncture five days rest for two days,a total of 4 weeks.Ovarian histomorphology was observed,and the protein expression of bcl-2,bax and p53 in ovary was detected.Results: 1.Morphological observation of ovarian tissue:In the normal group,there were different stages of follicle and corpus luteum and atresia follicle in the cortex.There was no distinct demarcation between the medulla and the cortex in the middle.There were many vessels,lymphatic vessels and nerves in the medulla,and there were no abnormal changes in the medulla.The model group could see the follicles at different stages of development in the cortex.Both the corpus luteum and the atresia follicle were not normal.The outer layer of the follicular membrane,the follicular membrane and the zona pellucida were not clear,the follicle cavity became smaller,no capillary was found,the medulla in the middle and the cortex had no distinct demarcation,no blood vessels,lymphatic vessels and nerves in the medulla,and the Chinese medicine group compared with the model group.At all levels,the follicles and luteal and atresia follicles were improved,the outer layer of follicular membrane,some parts of the follicular membrane and the boundaries of the hyaline zone changed,and the size of the follicle cavity in some parts was basically normal,even in the medulla,the blood vessels,lymphatic vessels and nerves were seen in the medulla,and the acupuncture group was not significantly different from the Chinese medicine group;the traditional Chinese medicine with acupuncture group was seen in the cortex.The follicles and corpus luteum and atresia follicles were recovered at different stages.The outer layer of follicular membrane,some parts of the follicular membrane and the boundaries of the hyaline zone were improved.The size of the follicle cavity was basically normal.The capillaries,medulla,and the boundary of cortex were still not obvious,and the blood vessels,lymphatic vessels and nerves in the medulla could be seen in most parts.2.Compared with the normal group,the expression of bcl-2 in the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly decreased(P <0.05);Compared with the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly increase(P <0.05).Compared with the traditional Chinese medicine group,the expression of acupuncture group had no significant statistical significance,and the expression of Chinese medicine with acupuncture group was significantly higher than that of acupuncture group significantly increased(P <0.05).Compared with the acupuncture group,the expression of Chinese medicine with acupuncture group increased significantly(P<0.05)3.Compared with the normal group,the expression of bax in the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly increased(P <0.05).Compared with the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly decline(P <0.05).Compared with the traditional Chinese medicine group,the expression of acupuncture group had no significant statistical significance,and the expression of Chinese medicine with acupuncture group was significantly lower than that of acupuncture group significantly decreased(P <0.05).Compared with the acupuncture group,the expression of Chinese medicine with acupuncture group decreased significantly(P<0.05)4.Compared with the normal group,the expression of p53 in the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly increased(P <0.05).Compared with the model group,the traditional Chinese medicine group,the acupuncture group,the traditional Chinese medicine with acupuncture group were significantly decline(P <0.05).Compared with the traditional Chinese medicine group,the expression of acupuncture group had no significant statistical significance,and the expression of Chinese medicine with acupuncture group was significantly lower than that of acupuncture group significantly decreased(P <0.05).Compared with the acupuncture group,the expression of Chinese medicine with acupuncture group decreased significantly(P<0.05)Conclusion: 1.Traditional Chinese Medicine and acupuncture of Tongbu Qijing therapy can effectively improve the survival of DOR rats and ovarian tissue morphology.2.Tongbu Qijing therapy combined with acupuncture and medicine to treat DOR can effectively improve the pathological state of rats,and its curative effect is better than using Chinese medicine or acupuncture alone.3.Traditional Chinese medicine compound and acupuncture of Tongbu Qijing therapy can effectively improve the pathological state of DOR rats,and its mechanism may be achieved through intervention of apoptosis of ovarian granulosa cells. |
