| Objective: To investigate the effect of mannose and pachymaran on human hepatocellular carcinoma cells(HCC)and to explore its potential clinic application for treatment of HCC patients.Methods: The cultured HepG2 cells were treated with different concentrations of mannose with/without cisplatin,the cell growth was monitored by CCK-8 assay.The cell scratch assay was used to trace the cell migration.Results:(1)The proliferation of Hep G2 cells gradually decreased with the increase of mannose concentration between 10-40 mmol/L and treated for 72 hours.Compared with the control group(glucose),the cell proliferation of mannose and cisplatin treatment were decreased,and the cell proliferation rate of mannose combined with cisplatin group was lower than that of cisplatin and mannose treated alone.(2)Mannose treatment changed morphology of Hep G2 cells as the cells showed separation,shedding and aggregation.(3)Mannose treatment reduced the migration rate of Hep G2 cells.(4)Pachymaran had no effect on Hep G2 proliferation at the tested concentration.Conclusion: Mannose has an inhibitory effect on the proliferation of Hep G2 cells and when treated together with cisplatin showed better anti-tumor effect than cisplatin alone.This effect of mannose suggests its clinic potential for future investigation. |
