| Purpose:To explore that whether 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside(TSG)is able to promote bone formation and has a preventive effect on postmenopausal osteoporosis(PMOP)of rats by activating the BMP-2/Smad1/Runx2/Osterix signal pathway.Materials and methods: 60 female SD rats with 3 months age were selected and randomly divided into 6 groups with 10 rats in each one,including: control group,model group(OVX),alendronate sodium group(AL),TSG in high dose group(HD),TSG in the medium dose group(MD),and TSG in the low dose group(LD).All but the control group were ovariectomized for double side to establish postmenopausal osteoporosis rat model.Gave rats medicine daily by gavage on the 7th day after surgery for 8 weeks.After the last gavage,measured the bone mineral density(BMD)of rats of each group by using double energy X-ray absorptiometry(DXA).Take the double-sided hind limb tibia after anesthesia.Took biomechanics detection for the tibia samples;Made pathological slices to observe the morphological structure of bone tissue by HE dyeing method;Detected the expression of 4 target pathway proteins BMP-2,Smad1,Runx2 and Osterix and their genes by using Immunohistochemistry(IHC),protein imprinting(western blot)and real-time fluorescence quantitative nucleic acid amplification detection(q PCR)technology.All obtained experimental data were analyzed by SPSS20.0 statistical software.Result:1.Bone density test results showed that: compared to the control group,the BMD value of the OVX group was significantly lower(P<0.01),indicating the success of PMOP model rat mold making.2.Bone stress testing showed: The elastic modulus of the HD group is higher than that of the OVX group(P 0.05).3.HE dyeing results showed: in OVX group,the trabecular was thinner,the spacing became larger and the percentage of trabecular area under the same field of view decreased significantly(P<0.01),but all these indicators of HD group were improved significantly compared to OVX group(P<0.05).4.The results of IHC,western blot and q PCR showed: The protein and m RNA expression content of BMP-2,Smad1,Runx2 and Osterix could be significantly increased in each TSG group(P<0.05).Conclusion:1.TSG can effectively prevent and control PMOP in OVX rats,wherein high,medium and low dose of TSG have a certain effect,and the effect increases with the concentration in a given range.2.TSG may promote bone formation by raising the expression of BMP-2/Smad1/Runx2/Osterix signal pathway.3.TSG may achieve the function of preventing PMOP through BMP-2/Smad1/Runx2/Osterix signal pathway. |
