Purpose:To observe the effect of Shenkang suppository on the kidney physical change and renal function in model rats with chronic renalfailure(CRF).To evaluate the effect of Shenkang suppository on the levels of immune factor such as tumor necrosis factor(TNF-?),?2-microglobulin(?2-MG)and the levels of urate translation(URAT1)and ATP-binding cassette superfamily G member 2(ABCG2)from the jejunum,researching the vital role they play during the progress of CRF,meanwhile,verifying the achievement of retention enema of Chinese Medicine aimed at CRF patient.Material and method:Fifty SD rats were divided into blank group(12rats)and model group(38rats)according to the random principle.The chronic renal failure model was made according to the Yokozawa method in model group.After successful modeling,they were randomly divided into model group,Shenkang suppository group,and Niaoduqing gavage group.Rats in Shenkang suppository group were given the Shenkang suppository through the anus.Rats in Niaoduqing gavage group were given Niaoduqing liquor through gavage.And the rats in the blank group and model group were given distilled water for intragastric administration.After 2weeks,The level of serum creatinine(Scr),blood urea nitrogen(BUN),serum albumin(ALB),TNF-?,?2-MG,and jejunum wasmeasured.The expression of URAT1 and ABCG2 was detected by immunohistochemistry.The data were analyzed using SSPS22.0.Results:1.When they were compared with the rats of blank group,the rats with blaek color of model move slowly and eat less,whose renal function release.The white renal under microscope shows fibrosis and broken kidney tubules.It shows that we finish the model making successfully.2.The level of Scr,BUN,UA,TNF-?and?2-MG in the rats with Shenkang suppo sitory group was lower than Niaoduqing gavage group and model group(P<0.05).SCr The model group 87.82±7.37?mol/L;Niaoduqing group 35.98±7.34?mol/L;Shenkang suppositorygroup30.48±3.63?mol/L,(P<0.01);BUN Model group19.76±2.45mmol/L;Niaoduqing group 10.48±1.89mmol/L;Shenkang suppository group 7.98±1.78mmol/L,(P<0.01);UA Model group 297.54±12.38mmol/L;Niaoduqing group 164.58±11.20mmol/L;Shenkang suppository group 112.75±10.57mmol/L,P<0.01)The influence of Shenkang suppository is lager than its o f Niaoduqing group(P<0.05).3.There was no significant difference among the ALB levels in the rats with Shenkang suppository group,Niaoduqing gavage group and the model group(P>0.05).4.The level of URAT1 on the jejunum tissues in the rats with Shenkang suppository group was lower than Niaoduqing gavage group and model group(P<0.05).5.There was no significant difference among the ABCG2 levels in the rats with Shenkang suppository group,.Niaoduqing gavage group and the model group(P>0.05).Conclusion:1.The Shenkang suppository could exert the effect of tonifying the bowels and relieving turbidity after absorption to protect kidney function by intestinal absorption,which was specifically exerted in three aspects:removing small and medium-sized molecules of urine toxin,reducing inflammatory factors and promoting the excretion of uric acid.2.Shenkang suppository can effectively release the level of Uric Acid by improving renal function.3.Shenkang suppository can effectively release the level of Uric Acid by controlling the expression of URAT1.4.Shenkang suppository has no effect on the expression of ABCG2 on the bowl.5.Shenkang suppository has no risk on nutrition of renal rats.
